Object i ve The purpose of this study was to study the effects of deep-sea fish oil on improving memory function through animal experiment and human experiment, in order to provide scientific evidence for its clinical application.Methods1.Safety toxicity studies of deep-sea fish oilMake clear the toxicity classification of deep-sea fish oil by transoral acute toxicity, and explore the potential mutagenic action by Ames test,mouse bone marrow micronucleus test and detection of chromosome aberration experiment, and further study the toxicity by 30-day feeding experiment. Summing up the above, make the safety evaluation of deep-sea fish oil.2.Effects of deep-sea fish oil on improving memory of mice240 male-mice weighted from 18 to 22g, were randomly divided into control group, low-dose group, middle-dose group and high-dose group of deep-sea fish oil. Four groups were separately treated with Og/kg-BW,0.15g/kg-BW,0.30g/kg-BW and 0.60g/kg·BW deep-sea fish oil daily by gastric perfusion for 30 days. The memory improvement effects of deep-sea fish oil were investigated by Step-down test, step-through test and water maze.3.Effects of deep-sea fish oil on improving memory of humanUsing self-control contrast and comparison test design between groups,200 subjects were selected and divided randomly into the intervention and control group. The intervention group was treated with taking orally 1.8g/60 kg·BW of deep-sea fish oil daily for 30 days. The effects of deep-sea fish oil on memory improvement were investigated by point to memory, associative learning, meaningless graphics to confess, graphics free memory and like features association.Results1 Safety toxicity studies of deep-sea fish oilTransoral acute toxicity:Maximum Tolerated Dose (MTD) are larger than 15.0g/kg·BW. Genetic toxicity tests:The results of Ames test,mouse bone marrow micronucleus test and detection of chromosome aberration experiment were negative. After treated rats with 0.75,1.50 and 3.00g/kg·BW deep-sea fish oil daily by gastric perfusion for 30 days, there were no statistically significant differences on body weight,weight increment,feed efficiency and dirty body ratio compared with control groups(P>0.05). The biochemical indexes and hematological indexes were normal, and anatomical saw no abnormal. No pathological and histological change was seen. No significant toxicity was seen by 30-day feeding experiment.2.Effects of deep-sea fish oil on improving memory of miceIn the middle-dose deep-sea fish oil group, the latency phase was prolonged in the Step-down test, and the final time was shorted in the water maze (P<0.05). No significant differences of the latency phase in the Step-down test and the final time in the water maze were observed among other groups (P>0.05).3.Effects of deep-sea fish oil on improving memory of humanAt baseline, no significant differences were observed in the subscales points, total points and MQ between intervention group and control group (P>0.05). In the control group, no significant differences were observed in the subscales points, total points and MQ before and after the trail (P>0.05). In the intervention group, the subscales points, total points and MQ increased significantly after the trail compared with those before the trail (P<0.01). After the trail, the subscales points, total points and MQ increased significantly in the intervention group than those in the control group (P<0.01). The results suggest that deep-sea fish oil could improve memory function in human according to the Standard of Health Food Test and Assess 2003Conclusions1.Deep-sea fish oil was at non-toxic toxicity level.2.Deep-sea fish oil has the effects on memory improvement in mice.3.Deep-sea fish oil has the effects on memory improvement in human.
