Abstract/Summary:
Objective:To investigate the expression of EV (including CVB),ADV and CAR in CRC, to study the relationship among EV(including CVB), ADV, CAR and the development of CRC, and analyze the clinicopathologic characteristics of CRCMethods:The expression of CAR in 100 cases of CRC and paraneoplastic epithelial tissue was examined using IHC. And the relationship among EV(including CVB) ADV, CAR expression and clinicopathologic was analyzed.The detection of EV(including CVB) and ADV in 100 cases of CRC and paraneoplastic epithelial tissue and cell lines CACO2 were developed using PCR/RT-PCR or cell culture.Results:1. Expression of CAR:All staining on the cell membrane. Compared with paraneoplastic epithelial tissue, CAR is down-regulated in tumor tissues. The down-regulation in rectal cancer was 26.31%, and in colon cancer 25.58%. Both showed statistical significance (P<0.05). The difference in hepatic metastasis, histological differentiation showed statistical significance (P<0.05). There were no statistical significance in age, sex, Dukes stage, tumor diameter, lymph node metastasis, pathology type (P>0.05). CAR expression in liver metastasis was more frequent than in primary CRC, but no statistical significance (P>0.05).2. Detection of EV or ADV:The EV RNA (CVB RNA) was detected in CRC (2%) and paraneoplastic epithelial tissue (2%), but there was no CPE through Vero cell culture. The EV RNA was undetected in cell line Caco-2, and there was no CPE through Vero cell culture. The difference of EV between CRC and paraneoplastic epithelial tissue shows no statistical significance.The ADV was detected in CRC (2%) and paraneoplastic epithelial tissue (3%), but there was no CPE through human Embryo Kidney 293 cell culture. The ADV DNA was undetected in cell line Caco-2, and there was no CPE through human embryo kidney 293 cell culture. The difference of ADV between CRC and paraneoplastic epithelial tissue shows no statistical significance.3. Correlation among EV, ADV and CAR:The difference of CAR with EV(+)/EV(-) or ADV(+)/ADV(-) shows no statistical significance(P>0.05). Conclusion:1. Compared with paraneoplastic epithelial tissue, CAR is down-regulated in tumor tissues, suggesting that the influence of CAR expression on therapeutic efficacy should be carefully considered when planning a regimen of gene therapy using adenovirus vector.The difference in hepatic metastasis, histological differentiation showed statistical significance (p<0.05), suggesting the relevance between CAR and malignant biological behaviour of CRC. There were no statistical significance in age, sex, Dukes stage, tumor diameter, lymph node metastasis, pathology type (p>0.05). CAR expression in liver metastasis was more frequent than in primary CRC, but no statistical significance (P>0.05).2. EV RNA or ADV DNA can be partly detected in CRC and paraneoplastic epithelial tissue. The difference of EV or ADV between CRC and paraneoplastic epithelial tissue shows no statistical significance.3. The down-regulation of CAR with EV(+)/EV(-) or ADV(+)/ADV(-) in CRC shows no correlation.
