OBJECTIVE:To observe the recovery of neurological function and investigate the protective mechanism, After mesenchymal stem cells(BMSCs) of vascular rats were transplanted by stereotaxis and vena caudalis path. This will provide therapy foundation in clinical MCSs application on vascular dementia and help to discover more effective therapy of vascular dementia.METHODS:1. Bone marrow tissues of rats were collected and theBMSCs Were separated with density gradient centrifugation and screening the cells adherent to the Plastic surface. The BMSCs were cultivated in specified medium..Before the transplantation, BMSCs were labeled by bromodeoxyuridine(BrdU).2. Wistar rats were randomly divided into 5 groups:control group; saline group; dementia model group; stereostatics graft group; tail vein grafts group.Two-vessel occlusion(2VO) was employed to make vascular dementia rat models. Stereotactic transplantation group and the tail vein graft rats 7 days after injury to bone marrow mesenchymal stem cells 1×106 months, by means of stereotactic transplantation Hippocampus, tail vein via the tail vein graft group, In the saline group, an equal volume of saline was utilized, In the control group, the rats were left intact.3. After modeling and the stem cell therapy are the Morris water maze behavioral test, including the place navigation test pilot and space exploration.30 days after stem cell transplantation the rats brains were removed to prepare paraffin sections, rows hematoxylin-eosin(HE) staining pathological changes. To evaluate the relationship between survivorship migration and differentiation,Brdu,NSE and GFAP were choosed as the markers of proliferation and differentiation. Brdu+NSE,Brdu+GFAP were identified by immunohistochemisty staining. To investigate The repair's mechanism of MCAO rats by analyzing the conditions of BMSCs'survivorship, migration,differentiation Through different transplanted path.RESULTS:(1) The results showed that by gradient centrifugation and adhesion culture,BMSCs could be isolated and culture-expanded.BMSCs Primary identification can be accomplished by flow cytometry. (2)Permanent bilateral common carotid artery ligation model of vascular dementia, through Morris water maze behavioral test, confirmed that successful model. (3)double-staining cells of immunohistochemistry could be found at the center of damage site and the surrounding site in rats of the transplantation group. (4) BMSCs transplantation group could be observed in the grafted sites Brdu ten NSE, Brdu+GEAP double staining positive cells, but failed to observe the neuronal precursor cells of the flag Doublecortin (DCX) from the lateral ventricle subventricular zone (SVZ) and the SVZ to the injury of the hippocampus rostral migratory stream (RMS) of the migration.CONELUSION:(1)BMSCs Can be isolated,passaged and proliferat; (2)BMSCs transplanted by me stereotaxis and vena caudalis Can survival and differert into neuron like cells and neuroglia like cells? and can improve neurological function.(3)Permanent bilateral common carotid artery ligation model of vascular dementia simple. Reliable, and can be used for study of vascular dementia. BMSCs transplanted bv me stereotaxis and vena caudalis Can survival and differert into neuron-like cells and neuroglia-like cells, and can improve neurological function.
