Objective Cervical cancer is a common malignancy among women, ranked second among in the world of women malignant tumors. The goal of this study was to analyze relationship between the expression levels of Caveolin-land Ki-67 and HPV infection in normal cervical tissue, cervical intraepithelial neoplasia (CIN) and cervical carcinoma, and to analyze its relationship with clinicopathological parameters of cervical cancer-related links to explore the above-mentioned target happened in cervical cancer development, invasion and metastasis, for the diagnosis and treatment of cervical cancer to provide a new and effective target.Methods Using streptavidin-biotin-protein-linked peroxidase method (ie,SP) immunohistochemical method to detect the expression of Caveolin-1 and Ki-67 and HPV16/18 infection in 15 cases of normal cervical tissue,35 cases of CIN tissue,50 cases of cervical cancer. Analysis of cervical cancer in Caveolin-1 and Ki-67 expression and clinical stage, histological grade and other clinicopathological parameters of relevance. At the same time the relationship between Caveolin-land Ki-67 and HPV16/18 infction was studyed in the development of cervical cancer for a preliminary study.Results 1. Caveolin-1 were expressed mainly in normal tissues and endothelisl cells, but also expressed in cancerous lesions; Ki-67 were expressed mainly in cancerous lesions, but also expressed in some normal tissues.2.Positive expression rate of Caveolin-1 in normal cervical tissues,CIN,cervical cancer tissues were 100.0%,77.14%,48.0%,the trend has gradually decreased (P <0.01). Among them,significant difference was not existed between normal cervical group and CIN group(P>0.0167), but there was significant difference between normal cervical group and cancer group(P<0.0167),also between CIN group and cancer group(P<0.0167).3.Positive expression rate of Ki-67 in normal cervical tissues,CIN,cervical cancer tissues were20.0%,57.14%,86.0%, the trend has gradually increased (P<0.01). Among them, significant difference was not existed between nonnal cervical group and CIN group(P<0.0167), but there was significant difference between normal cervical group and cancer group(P<0.0167), CINgroup and cancer group(P<0.0167).4. In cervical carcinoma, positive rate of Caveolin-1 protein at FIGO stagesⅡb-Ⅳperiod was lower than theⅠ-Ⅱaperiod (χ2=5.265,P<0.05), the low degree of differentiation group was lower than that high degree of differentiation group(χ2=5.467,P<0.05), with lymph node metastasis group was lower than that without lymph node metastasis group (χ2=3.907,P<0.05); Expression of Caveolin-1 in cervical cancer were not related with age,gross type,histological types (P> 0.05).5. In cervical cancer, positive rate of Ki67 protein at FIGO stagesⅡb-Ⅳaperiod was higher than theⅠ-Ⅱaperiod (χ2=4.948,P<0.05),the low degree of differentiation group was higher than that high degree of differentiation group(χ2=4.060,P<0.05), Expression of Ki67 in cervical cancer were not related with age,gross type,lymph node metastasis and histological types(P>0.05).6.Positive expression rate of HPV16/18 in normal cervical tissues,CIN,cervical cancer tissues were 13.33%,48.57%,92.0%,the trend has gradually increased (P<0.01). Among them,significant difference was not existed between normal cervical group and CIN group(P>0.0167), but there was significant difference between normal cervical group and cancer group(P<0.0167), CIN group and cancer group(P<0.0167) Expression of HPV16/18 in cervical cancer were not related with age,degree of differentiation,FIGO stages,gross type,histological types and lymph node metastasis (P<0.05)7.The results of correlation study on expression of Caveolin-1 and Ki-67 show a negative correlation in cervical carcinoma(rs=-0.684,P<0.05).The relationship between Caveolin-1 and HPV was negative correlated(rs=-0.752,P<0.05);The relationship between Ki-67 and HPV was positively correlated(rs=0.658,P<0.05)。Conclusion Expression of Caveolin-1 in cervical carcinoma was decreased,which was related to development of cervical cancer. Expression of Caveolin-1 in cervical cancer were related with FIGO stages,degree of differentiation and lymph node metastasis.With the increasing of FIGO stages,decreasing of degree of differentiation and with lymph node metastasis expression of Caveolin-1 in cervical carcinoma was decreased, which was related to malignant degree of cervical cancer. Expression of Ki-67 reflect proliferation activity of cell in carcinogenesis of cervical epithelial, which was in contast with expression of Caveolin-1.Abnormal expression of Caveolin-land Ki-67 in cervical carcinoma were relate to HPV16/18 infection. Combined detection of Caveolin-1 and Ki67 and HPV16/18 may be a new target of cancer diagnosis and treatment and a new way to assess the severity of cervical cancer.
