Abstract/Summary:
OBJECTIVE:1,In order to research the mechanism of VM and anti-tumor therapy in view of VM. we build the Model of 3-dimentional culture in vitro of Choroid melanoma cell line OCM-1.2,This research observed the effects of Cur on Vasculogenic mimicry and PI3K way in Choroid melanoma cell line OCM-1 in vitro 3-dimentional culture. We provide the part of experiment for the anti-tumor therapy and the mechanism of Cur.METHODS:1,Put OCM-1 cells in vitro 3-dimentinal culture, Observe VM and Stained with PAS.2,Put OCM-1 cells in vitro 3-dimentinal culture,observed if it could form vasculogenic mimicry and the influence of different density(0umol/L 10umol/L,20umol/L,30umol/L,40umol/L) Cur in OCM-1 cell line. Stained with PAS and immunohischemistry tested the PI3K and EphA2 expression of 3-dimentional cultured OCM-1.3,All the information were put into software of SPSS 16.0 to analysis.RESULTS:1,Choroid melanoma cell line OCM-1 could form vasculogenic mimicry in vitro 3-demintional culture. During 3-dimintionaI culture, OCM-1 cells stretched out thin and long apophyses at the fourth day, and linked each other to form wreath and net-work structure at the seventh day. Tumor cells were separated from the tubes by PAS-positive matter like basement membrane.2,The morphology observes for the influence of Cur to form VM of OCM-1 cell Cur markedly inhibited VM and dependenced M-concentration of Cur.3,Immunohischemistry tested the PI3K and EphA2 expression of 3-dimentional cultured OCM-1:Cur markedly inhibited VM and expression of PI3K and EphA2 (P<0.05). CONCLUSION:1,Choroid melanoma cell line OCM-1 could form vasculogenic mimicry in vitro 3-demintional culture, we build the Model for research the mechanism of VM and anti-tumor therapy in view of VM.2,Cur markedly inhibited VM and one of the mechanism was probably throughdecreasing PI3K and EphA2 expressions.3,The anti-tumor mechanism of Cur can be probably through the phosphatidylinositol-3-kinase (PI3K) signaling pathway and meantime inhibite angiogenesis of tumor.
