Objective and Background:The bladder cancer is one of the most common and hazard diseases in the malignant tumor of urinary system in china. The molecular mechanism of the development and the progress of the bladder cancer is poorly understood. Doctors are still in lack of the effective diagnosis and prognosis tumor marker. There have a few reports on the correlation between the expressions of CD147 and MDM2 gene in bladder cancer. This study was to discovery the expressions and significance of CD147 and MDM2 gene in both bladder cancer and the normal bladder tissue. We would analyzed the relationship between CD147 and MDM2 gene and development, clinical stage, and pathological grade of bladder cancer, willing to provide new clinical theory basis for the diagnosis and therapy of bladder cancer.Material and Methods:This study included 42 cases of patients, from September,2008 to Febrary,2009, who admitted in the General Hospital of Tianjin Medical University for the treatment of bladder cancer.8 specimens of normal bladder tissues were acquired as control. Immunohistochemical staining for was carried out and SPSS 16.0 for windows was used to evaluate the association between CD147 and MDM2 expression and development, progress, clinico-pathologic parameters.Results:1. The expressions of CD147 and MDM2 protein were over expressed with the positive staining rate being 69.0% and 40.5%, While none of CD147 and MDM2 expression were found in normal bladder tissue. Both of them were significant differential between bladder cancers and normal bladder tissues.2. The different degree of CD147 expression was related to clinical stage, and pathological grade of bladder cancer. The positive rate of CD147 expression in the invasion tumor was 73.1%, while the superficial tumor was 62.5%. A statistically significant difference was observed between them. The positive rate of CD147 expression in the high-grade bladder tumor was 100%, while the low-grade bladder tumor was 85.0%. A statistically significant difference was observed between them. CD 147 expression in the non-recurred group was 63.1%,while the recurred group was 73.5%. However, No statistically difference between them.3. The different degree of MDM2 expression was related to clinical stage, and pathological grade of bladder cancer. The positive rate of MDM2 expression in the invasion tumor was 62.5%, while the superficial tumor was 73.1%. The positive rate of MDM2 expression in the high-grade bladder tumor was 25.0%, while the low-grade bladder tumor was 46.7%. MDM2 expression in the non-recurred group was 57.9%, while the recurred group was 23.5%. A statistically difference between them.4. Combined application of CD147 and MDM2 positive expression rate was 80.9%.The clinical and pathologic stage grading and without recurrence after without related.Conclusion:1. The study had shown that there was more expression of CD147 and MDM2 gene in bladder cancer than in the normal tissue. A statistically significant association was observed between them.2. CD147 have rlevance to infiltrating depth and lymph node metastasis of bladder tumor. The high expression of CD147 gene, the worse clinical-pathology features. Which show CD 147 is one of high level invasive bladder cancer markers.3. The low expression of MDM2, the worse clinical-pathology features. Which show MDM2 is one of low level the infiltration of bladder markers.4. CD147 and MDM2 gene have close relationships with the prognosis of bladder cancer. they were the objective marks to estimate the behaviors of bladder tumor.
