Objective:Toll receptors (TLRs), as a family of pattern recognition receptor,can discern related pathogen molecular in cell surface. It plays an important role in respiratory innate immunity and acquired immunity. the defect expression or poor response of TLRs often leads to chronic respiratory infection. Actl is the important signal molecule in IL-25 signal pathways, promoting Th2 inflammation; A20, as a potential inhibitor in NF-κB signaling pathway, was encoded by immediate early response gene. this study was to applicate pathogenic Escherichia coli fimH gene (TLR4 natural ligand) into the vaccine PPA prepared by Pseudomonas aeruginosa, interventing allergen-induced or virus-infected bronchial epithelial cells and observing the effect on respiratory epithelium PPA,Actl,A20 and proliferation function to find TLR4 for respiratory hyperresponsiveness diseases.Methods:for HBECs, stimulated by OVA or infected by RSV, then through PPA intervention, using flow cytometry to detect the cell cycle HBECs.adopting fluorescence quantitative PCR to detect the mRNA-expression differences of Actl and A20.Result:1,cell cycle:PPA can increase the numbers of S-phase cells in HBECs whatever treated by OVA or by RSV (P<0.05).2, TLR4: PPA can significantly increase mRNA's expression in HBECs TLR4 treated by OVA(P<0.01); and PPA increased mRNA's expression in HBECs TLR4 treated by RSV (P<0.05).3,Act1:PPA can significantly decrease mRNA's expression in HBECs Actl treated by OVA(P<0.01); while PPA increased mRNA's expression in HBECs Actl treated by RSV (P<0.01).4,A20:PPA can significantly increase mRNA's expression in HBECs A20 whatever treated by OVA or by RSV (P<0.01).Conclusion:1,PPA can significantly promote the proliferation of human bronchial epithelial cells and can interfere with OVA or RSV in inhibiting the proliferation of epithelial.2,PPA intervention can increase the expression of TLR4 Of human bronchial epithelial cells.3,PPA intervention can inhibit the allergen -induced expression of Actl in human bronchial epithelial cells.4,PPA intervention can increase the expression of A20 of human bronchial epithelial cells, inhibiting inflammation response induced by NF-κB pathway.
