In this paper, we initial studied the impact of ractopamine hydrochloride in chicken serum glucose and blood lipid content, the residue depletion of ractopamine in chicken serum and tissues were being studied by both liquid chromatography-mass spectrometry technology and enzyme-linked immunosorbent analysis techniques. Then we made a comparative analysis on the results between the two methods.Eighty healthy three-yellow chickens were randomly divided into four groups, the experimental group (n=24), normal control group of eight, from the age of29days on.both experiment groups of three-yellow chickens were fed diet added1or3and9mg/kg RAC for21days, the normal control group did not add. The cholesterol, triglycerides and serum glucose levels in the collected blood sample from each goup was detected atld,2d,3d,4d,7d,14d,21d after feeding RAC. the results showed as followed:there were some impact on the three-yellow chickens serum cholesterol, triglycerides and serum glucose levels change which fed diet added RAC. the content of the experiment group were reflected in the fluctuations more than the control group, the long-term administration can reduce cholesterol and serum glucose levels in chicken serum on different degrees and little effect on triglvcerides.The RAC on serum and tissue samples were detected at1d,3d,7d,14d,21d after treatment by two analysis methods and ld,3d,7d withdrawing drugs. The results showed that:the two methods measured results have good consistency.the consistency of the detection results in muscle and stomach samples were better than in serum and liver. Overall, the residual concentration in different tissue was liver> stomach> muscle> serum during the feeding period. While the residual concentrations from high to low at withdrawing drugs was stomach> liver> muscle> serum.The above studies have shown that there were some impact of ractopamine hydrochloride added in diet on chicken serum glucose and blood-lipid content, through21days of continuous medication, the serum cholesterol and serum glucose levels in experiment group were lower than the normal control group and there was no significant difference in triglyceride content. We could draw the conclusion that RAC was easily remained in tissues of the three-yellow chickens, the residual concentration and the velocity of RAC metabolization in different tissues were different both. And there have a good consistency by using LC-MS and ELISA to detect the tissues and serum sample which contained RAC.
