| This study was determined to predict all the metabolisms of Volvariella volvacea withbioinformatics analysis under the premise of the Chinese straw mushroom genome sequenced byour laboratory. Literature review to find the metabolic pathways of indole-3-acetic (IAA)biosynthesis pathway build and the IAA metabolism in V. volvacea was build. KAAS onlinesoftware in KEGG website was used to predict of V. volvacea metabolism with the genomic data.The total number of metabolic pathways was predicted to236, including44genes involved in thepathway of tryptophan metabolism, including the15IAA metabolism-related genes. These geneswere involved in IAA biosynthesis in three ways:(1) the indole-3-acetamide (IAM) pathway;(2)the tryptamine pathway;(3) the indole-3-acetaldoxime (IAOx) pathway.Primer pairs were redesigned to amplify and sequence the IAA metabolism-related genes inorder to verify the accuracy of the V. volvacea genome splicing data. Protein domain search, thesearch of homologous proteins in phylogenetic tree construction and protein conserved motifs, thethree aspects of the analysis is further confirmed by the data reliability of the coding gene. Inaddition, these genes were classified with the bioinformatics analysis.In this study, the3'-tag digital gene expression (DGE) of Volvariella volvacea was performedto analyze the differential expression of IAA metabolism-related genes in fruiting developmentalstages from V. volvacea. There are21IAA metabolism-related genes which were significantlydifferentially expressed in differential stage from V. volvacea. Six genes were selected randomly(each enzyme contained1-2genes) to study theirs expression by QPCR. QPCR results support theDGE data.IAA concentration was determined by HPLC at different developmental stages of fruitingbody stipe from V. volvacea. It contains the highest IAA concentration in the button stage of V.volvacea, which can reach0.7875mg/kg with significant differences.V. volvacea mycelium growth rate was detected by adding different concentrations ofexogenous IAA solution in on PDA medium. The data demonstrated that high concentrations ofIAA solution would inhibit V. volvacea mycelium growth, while low concentrations has no effecton mushrooms. Test was made by spraying different concentrations of exogenous IAA solution onthe bed surface (after the day of spraying water on the bed surface). The result shows that sprayingIAA solution with10-5mol/L concentration will promote the growth of primordial from the V.volvacea, increase the fruiting higher yield, and its parachute time is earlier than that in control.Test was made by soaking the postharvest mushrooms with different concentrations of exogenousIAA solution18hours. The result displays that the stipe of V. volvacea will continue to grow andthe length of the longest is grown in IAA solution with10-8mol/L concentration. But in general,on mushrooms continue to grow with no significant differences. |
