In this study,'Jintian Huangjia Seedless' Grape is used as female parent.'Centennial Seedless','Fantasy Seedless' and 'Crimson Seedless' Grape is used asmale parent. The average of fruit weight, seed numbers, seed average weight, verticaland horizontal size and embryo morphology are investigated. From40days to85days after the pollination, the hybridized and the self-pollinated embryos werecultured in different media by embryo culturing combining beak cutting to get rid ofthe dormancy. Studies of the hybrid breeding and the culturing method were carriedon during the process of developing and germinating of embryos.The result indicated that:1. The best sampling time for embryo development of the grape 'JintianHuangjia Seedless' by self-pollinating is85days after pollination. Embryos rate,germination rate and seedling rate is55.78%,23.08%and11.54%respectively. Thebest sampling time for embryo development of the grape 'Jintian Huangjia Seedless'×'Centennia Seedless' is70days after pollination, the grape 'Jintian HuangjiaSeedless'×'Fantasy Seedless' is65days,the grape 'Jintian Huangjia Seedless'×'Crimson Seedless' is85days. Seedling rate is7.50%,7.69%and11.76%respectively.2. The suitable medium of embryo resuce is Improved N+6-BA2.5mg·L-1+IAA1.5mg·L-1+AC0.5g·L-1+Pro2.0mmol·L-1, Improved N+6-BA2.5mg·L-1+IAA1.5mg·L-1+CH500mg·L-1, Improved N+IBA2.0mg·L-1+6-BA0.5mg·L-1+GA30.5mg·L-1+AC1.5g·L-1and Improved N+6-BA0.5mg·L-1+IAA1.0mg·L-1+AC0.5g·L-1+CH500mg·L-1respectively for the grape 'Jintian HuangjiaSeedless' by self-pollinating,'Jintian Huangjia Seedless'×'Centennial Seedless','Jintian Huangjia Seedless'×'Fantasy Seedless' and 'Jintian Huangjia Seedless'×'Crimson Seedless'. It is essential to add6%sucrose, the pH value of5.0solidmedium is used, cultured under the conditions of25±1℃,2000Lx.3. The appropriate subculture medium of embryo rescue seedlings is the1/2MS+NAA0.1mg·L-1+20(25)mg·L-1Agar. Plant regeneration efficiency is the highest and its stem is thicker, its leaves are larger and dart green, and internodes areshorter.4. The appropriate rejuvenation medium of embryo rescue seedlings is MS+IBA0.02mg·L-1+PP3331.0mg·L-1or CCC1.5mg·L-1. With the media the seedlingheight is3.43cm and1.72cm respectively. And the stem is1.25mm and0.92mm,which are suitable for grafting of the test-tube seedings.5. Good quality plants cultured for50days were Selected. Tube seedlings haveto exercise with suitable temperature and humidity by degrees before transplantation,which make them adapt to national circumstances. Pruned root and transplanted into2/5vermiculite (upper layer) and3/5nutrient soil(under layer)the nourishing soil, theplants survive.
