| There is a production and drinking history of "Kudingcha" for at least more than2000years in China. Of all the species called "Kudingcha" Ilex kudingcha C. J. Tseng is the most influential one in the the cullivation area, product variety, yield, price, as well as the output value. The products of I.Kudingcha have been exported to more than a dozen countries and regions.Been a newly-reported disease Colletotrichum illics-kudingchae has been the most serious disease on the yield and on the product quality of I. Kudingcha. To breed the resistant variety is one of the effective measures for controlling the diseases of crops. However, there are a series of of problems (such as time-consuming, laborious, the low choose efficiency and so on) in the traditional cross breeding. An effective method for breeding resistant variety is to establish a technical system for the rapid detection of plant diseases. A dual molecular marker technique, i.e. RAPD-SCAR (Random Amplified Polymorphic DNA-Sequence Characterized Amplified Region) can be used to identify rapidly the resistance of the breeding materials, to screen conserve the valuable germplasm materials, and to eliminate these materials whith aren't in keeping with the breeding targets, so that the breeding workers could shorten the breed period, heighten the choosing efficiency, and speed up the breeding process.In order to develop the specifical SCAR marker closely linked to the resistant gene Colletotrichum illics-kudingchae in Ilex kudingcha C. J, the different germplasm materials which the resistibility to Colletotrichum illics-kudingchae were konwnly high-resistance/high-senstive,were at first analysed via RAPD molecular marker with92random primers. And from which a few specifical RAPD marker closely linked to the resistance gene found out(including these specifical fragments: S69-300, S227-320, S227-2073and S247-431).Then these specifical fragments were retrieved, amplified and sequenced, and12pairs of specific SCAR primers were designed based on the sequenced results. And then these newly designed SCAR primers were used to convert the specifical RAPD markers into SCAR primers. At present2pairs of specifical SCAR marker have been successfully used to convert the specifical RAPD markers into SCAR marker. The resistibility to Colletotrichum illics-kudingchae in85germplasm materials of Ilex kudingcha C. J. Tseng were determined with these2pairs of SCAR primers. The results showed that the experimental results of SCAR marker were fundamentally in keeping with the observational results of the field, and with that of artificially inoculation.Their coincidence percentages were as high as96.47%. |
