Fructan is a kind of polymer of fructose molecules synthetized by fructosytranses(FTs) with the substrate of sucrose. Fructan as a main kind of storage carbohydrate exits in about15%of angiosperms. Besides that, Fructan also has many other important physiological functions, such as osmotic adjustment to prevent cell deydration and mantain the stability of the membrane under drought, high salt and other abiotic stress. Plants response to environmental stresses through fructan metabolism, plant resistance to various environmental stresses can be improved by accumulation of fructan. The1-SST gene of Jerusalem artichoke had been transferred into Reyan7-33-97by the method of Particle Bombardment and Agrobacterium-mediated transformation, expecting to acquire thel-SST-transgenic rubber plant and cultivate stress-resistance varieties of the rubber.In this paper, we compared anthers, embryoids and long-term subculture proliferation anther callus of Reyan7-33-97in aspects of callus culture, embryoid induction and seeding emergence. The results show that embryoids are better than the other two in embryoid induction and seeding emergence, subculture proliferation can indirectly proliferate embryoid. Embryoid of subculture proliferation can still fast be induced into embryos, and is suitable for long-term proliferation and saving.Ten resistant transgenic seedlings had been obtained through Particle Bombardment transformation of Reyan7-33-97and1.5mg/L glufosinate selection. Two positive seedlings were verified by PCR and RT-PCR, further more, Southern blot also showed a peculiar objective band. All above showed the1-SST gene has been successfully integrated into the genome DNA of the rubber.Four resistant transgenic seedlings had been obtained through Agrobacterium-mediated transformation of Reyan7-33-97using suspension co-culture and1.5mg/L glufosinate for selection. Two positive seedlings were verified by PCR, only one positive seedlings by RT-PCR. There is also no obvious objective band by Southern blot hybridization. |
