The cadmium (Cd) content of Phaseolus vulgaris seedlings using atomic absorption spectrophotometric(FAAS) were determined in this study , and the content of malondialdehyde(MDA),soluble protein, Pro, and GSH et.al were also investaged using ultraviolet absorption spectrophotometry (UV-VIS) to study the effect of Cd on the growth of Phaseolus vulgaris seedlings in the hydroponic condition. The effects of Cd stress on DNA polymorphism in leaves of Phaseolus vulgaris seedlings were studied in this hydroponic experiments by using random amplified polymorphic DNA (RAPD) technique;Moreover,In this pot-soil culture experiment, physiological indexes such as glutathione (GSH), the phytochelatins (PCs), et al and hyperchromic effect of DNA were studied under the interactive treatment of cadmium and sulfur on seedling leaves of Phaseolus vulgaris. It is provided a theoretical basis for Phaseolus vulgaris products. The results in this study are as follows:The results showed that after 7 days of 0,20,40 and 80 CdCl2 mg·L-1 stress, the malondialdehyde(MDA)and cadmium content ascended ;the biomass and the total soluble protein content, Pro content and GSH content increased at low concentration conditions(20 CdCl2 mg·L-1)and then decreased in middle and high density condition(40 and 80 CdCl2 mg·L-1).A total of eight oligonucleotide primers (10bp) were found to produce unique polymorphic band patterns, and subsequently used to produce 46 bands of in the normal leaves of Phaseolus vulgaris seedlings. The RAPD profiles showed obvious differences between normal and exposed plantlets, and the change effect was dose-dependent.The genomic template stability was significantly affected by Cd pollution, and the DNA polymorphisms detected by RAPD technique could be used as an analytic approach for detecting the genotoxic effects of Cd stress on plants.The results showed that the contents of MDA and hyperchromic exhibited a significant increase with the rise of treatment concentration of cadmium, but they decreased significantly after treatment simultaneously with sulfur of 100mg·kg-1. Biomass of Phaseolus vulgaris treated with sulfur of 100mg·kg-1 ascended up more greatly than control.After more sulfur added in, acid-soluble thiols (TAST),glutathione (GSH),the phytochelatins (PCs) showed an ascending trend.It is suggested that addition of sulfur in soil of light Cd pollution ameliorate Cd toxicity on Phaseolus vulgaris.
