Lamiophlomisrotata(Benth.)Kudo,is only one species of Labiatae.It is a peren- nial herb, is one commonly used herbal folk of Tibetan, Mongolian, Naxi and others, the whole plant as medicine,with high medicinal value. In this experiment, young leaves, petioles, young roots of the seed germination of wild Kudo seedlings with true leaves as explants. Observation of its callus induction, subculture, differentiation and morphogenesis, systematic analysis the internal mechanism of the dedifferentiation and differentiation. The purpose provide the theoretical basis and practice is through in vitro tissue culture by the rapid asexual reproduction, use of secondary metabolites, somaclonal variation, breeding and other aspects of research.The main conelusions on Lamiophlomisrotata (Benth.) Kudo as followed:1. MS medium as basic medium, MS+2,4-D1.0mg·L-1+6-BA0.5mg·L-1+NAA0.5 mg·L-1 as the induction medium.Callus induction rate can reach 80.0%, 70.0% and 91.3% respectively of young leaves, petioles and young roots as explants.2.Subculture when the selection of the same concentration of 2,4-D, NAA and 6-BA, Kudo callus subculture on MS are better than B5, N6 and MA.The best subculture medium is MS+2 ,4-D1.0mg·L-1 +6-BA0.5mg·L-1+NAA0.5mg·L-1+Inositol 100 mg·L-1+CH300mg·L-1+3%sucrose; Callus growth was "S" curve trend of dynamic change, the best time of subculture is the 25th,light culture is useful in callus growth; suitable temperature of subculture callus is 22℃.3. To add 1.0 mg·L-16-BA and 0.5mg·L-1NAA in the MS medium as differentiation medium, differentiation rates of different types callus is different, adventitious buds up to 85%; through the method of micro-Branch cutting propagation of adventitious buds was up to 5 times. MS0 medium induced the most significant rooting, rooting rate is 100%; eventually after acclimatization and transplanting, the survival rate is up to 90%.4. The best medium is MS+2,4-D2.0mg·L-1+6-BA0.5mg·L-1+ NAA0.5mg·L-1; 2,4-D for somatic embryogenesis on the independence embryogenic callus formation is necessary,to raise the concentration of 2,4-D is conducive to the occurrence of somatic embryos; sucrose concentration of 3% is also conducive to the occurrence of Kudo embryogenesis; Content of CH can influence the time of embryoid induction and formation.5.There were organogenesis and somatic embryogenesis on morphogenesis of Lamio- phlomisrotata (Benth.) Kudo. Kudo callus buds mostly foreign origin, but there are also the origin.It was same to other dicotyledom, they all experience the stages of proembryoid,globular embryo,heart-shaped embryo,torpedo embryo and cotyledom embryo.We assume that the origion of somatic embryo of Lamiophlomisrotata(Benth.) Kudo is from the single-cell. Both within the original cells of embryonic origin, but also foreign origin.
