L-ascorbic acid (AsA) which is generally called Vitamin C plays essential roles in maintaining human health and improving plant biochemistry reaction, especially when plants are in adversity stress. Studies show that IDH gene is a key enzyme in the pathway of AsA degradation. After analysis, we found that SDH gene may have the similar function with IDH gene and may be involved in ASA degradation. The high SDH activity may contribute to the low AsA content in apple trees and other Rosaceae trees. To further verify this deduction, In this study, we cloned IDH gene from Actinidia deliciosa cv.'Qinmei',and constructed plant express vectors of IDH and SDH1 genes, which lay the foundation for the further study on the relationship of these two genes and their relationship with AsA. The main results in this study are as follows:(1) cDNA overall length of IDH gene from kiwi fruit was obtained. The cDNA sequence length is 1239bp which has a whole open reading frame of 1080bp and encodes 359 amino acid. Blast analysis showed that apple SDH1 gene has high homology with kiwifruit IDH gene, which may support the above mentioned deduction;(2) Plant excessive express vector pWR-IDH was constructed and transformed into EHA105 successfully;(3) Plant excessive express vector pWR-SDH1 was constructed and transformed into EHA105 successfully;(4) Tomato leaves were transformed with pWR-SDH1 by Agrobacterium tumefaciens-mediated transformation. Transgenic resistant plants proved by PCR had been obtained;(5) Tomato leaves were transformed with pWR-IDH by Agrobacterium tumefaciens-mediated transformation. Transgenic resistant plants proved by PCR had been obtained;(6) Apple leaves were transformed with pWR-IDH by Agrobacterium tumefaciens-mediated transformation. Transgenic resistant plants proved by PCR had been obtained.
