| Myopalladin (MYPN) is a multifunctional protein whose main role is to maintain sarcomeric integrity and regulate Z-line structure, which function as scaffolds that regulate actin organization. Acyl-coenzyme A oxidase 1 (ACOX1) is the first and rate-limiting enzyme in peroxisomal fatty acid ?-oxidation and plays a key role in fatty acid metabolism and fat deposition. They are an important candidate gene for meat quality selection through marker-assisted selection (MAS).In our study, In order to produce scientific proof for marker-assisted selection of meat quality traits of chinese yellow cattle and built scientific basis for high-yield breeding skills of new breed, DNA and protein sequences of these genes related to meat quality traits were analyzed by biological software and bioinformatics. While the polymorphism of these genes were analyzed by bioinformatics, PCR-RFLP and PCR-SSCP method in Qinchuan (QC), Luxi (LX), Nanyang (NY), Jiaxian Red (JR), Xianan (XN), Xuelong Black(XL) and Simmental and Luxi crossbred steers (SL). The frequency, equitability, total heritability and relevance of meat quality traits of 2 SNPs were analyzed by Bio-statistics. The main resules of our study are as follwed:1. The characteristics and functions of gene sequences and amino acid information in MYPN and ACOX1 genes were predicted and analyzed by bioinformatics. The secondary and tertiary structure, signal peptide sequence, transmembrane region and functional domain of these 2 proteins were predicted and analyzed, too. The phylogenetic trees were also built by comparison with other species and conservative analysis of corresponding proteins in other species.2. PCR-RFLP technology were adopted to detect the polymorphism of one locus of MYPN gene, identify genotypes in bovine population and anaylze their association with meat quality traits, the results show as below: There are significant differences between the different genotypes for LEA (P<0.05) and WHC (P<0.01) . Animals with GG genotype have higher mean values of LEA and WHC than those with the AG and GG genotypes. No other significant correlation was observed between any of the marker genotypes at A1795G for the other traits.3. PCR-SSCP technology were adopted to detect the polymorphism of one locus of ACOX1 gene, identify genotypes in bovine population and anaylze their association with meat quality traits, the results show as below: there are significant effects on the UBF and UMAR (P<0.05), where animals with the genotype AA have higher mean values of UBF and UMAR than those with genotypes AC or CC. Besides, we also analyzed the association between genotypes and meat quality traits, including BFT, LEA, MAR, WHC, and TD, in another 71 QC individuals, there are significant differences between the different genotypes for BFT (P < 0.05), but we did not find any association between genotypes and MAR phenotypic data. |
