Tricholoma mongolicum Imai grows on Bayanbulak Grassland in Xinjiang Province, As it have the character of hypertrophy context, fresh and tender texture and good taste, it is a kind of precious edible and officinal mushroom and valuable resource in our country.Because of the fact that Tricholoma mongolicum Imai haven't been domesticated, besides the entironment where it lives has been destroyed,the number of wild Tricholoma mongolicum Imai is decreasing gradually,even facing the survival crisis and extinction.In this article, first selected strains J-4 was identified by the way of molecular biology, second studied on its biological characteristics, then optimized the strain medium and original medium of Tricholoma mongolicum Imai, in the end, studied on the effect of different raw material on the growth of Tricholoma mongolicum Imai, selected the proper formula, provided reliable reference of artificial domestication by building mathematical model.The main conclusions are following:1. Selected strains J-4 was identified by the way of ITS series analysis, The results showed that it shares 99% dentity with Tricholoma mongolicum, meanwhile constructing evolutionary tree to preliminary analyse phylogenetic relationships of Tricholoma mongolicum Imai.2. Study on the effect of differect carbon source,nitrogen source, mineral matter, vitamin and plant growth regulator on growth of Tricholoma mongolicum Imai. The results show that Tricholoma mongolicum Imai can use kinds of monose, disaccharide and polysaccharide, soluble starch is the best best carbon sources; All the organic,inorganic sources and amino acids investigated improved growth with beef extract being the best; 0.6 g/L nitrate of lime and lemery salt, 2 mg/L vitamin B6 promoted good mycelial growth. Among the phytohormones tested, 20 mg/L 6-BA and KT stimulated mycelial growth.3. The effects of four environment factors on the growth of Tricholoma mongolicum Imai mycelium were studied. The results show that the optimum pH is 5.7,the suitable temperature is 21℃, the optimum water content of medium is 64%, light has non significant action to mycelium growth.4. Experiments were performed to determine effects of supplementation of soluble starch with maltose, beef extract and yeast extract on growth of Tricholoma mongolicum Imai mycelium using quadratic regression orthogonal design, quadratic polynomial equation was developed to show the effects of each cultivation materials composition for Tricholoma mongolicum Imai mycelium, select the optimal formula for it. The results showed that the four materials can promote the growth rate of Tricholoma mongolicum Imai mycelium dramatically, and soluble starch was the best carbon source. The model estimated that, the best medium formula can be obtained when the concentration of soluble starch, maltose, beef extract and yeast extract were 18g/L, 8.55 g/L, 1 g/L and 0.6g/L, respectively.5. Effect of turf, bran and spent mushroom substrates to the growth of Tricholoma mongolicum Imai mycelium were studied using proportional mixture design and quadric quadratic regression orthogonal methods. The corresponding mathematical model was established. The results showed that augmentation in growth rate of mycelium was associated with an increase in content of bran, and spent mushroom substrates can be conducive to Tricholoma mongolicum Imai mycelium to some extent. The model estimated that, the best medium formula can be obtained when the concentration of wheat bran, spent mushroom substrates and turf were 11%, 14% and 75%, respectively.
