Study On Form Mechanism Of Fibrosis Capsule Wall Around Hepatichydaid

Object:After E.G invaded into human and sheep body, it will form focus of infection-Cy,In the evolution, E.G erosion the liver, there will be appearance of fibrous structure, the part near to E.G called capsula externa and the part near to liver called adventitia.we want to find changes of human and sheep liver DNA on genome level.1. we want to find different gene sites between nomal liver and Pathological tissues.2. By Laser microdissection and FP,hope to find change on genome level.Methods:1. By RAPD anlysis,find changes of polymorphism sites2. Through Cloning,Prepare specificity probe and differentional gene on proteomics.3. Get different issues and different periods cells by Laser microdissection.4. Get FP through Radiolabeled and Fluorescence Technology.Results:1. Because of specificity of disease liver,we improved the methods of extracting DNA and got one suitable method,which include increasing gringing force,addingultrasound,ectending processing time,and so on.Last we use Liquid nitrogen and Quartz sand to grind,use ultrasound to broke cells,and get one good result.2. Through simple experiments,difined reaction conditions of RAPD and got 10 good map of human and sheep liver issues.3. We got 4 clone of specificity probe by RAPD data analysis and cloning technology.4. By Laser microdissection and micro DNA extraction,we got 13 cell samples of different periods.5. Using Radiolabeled Technology,we got 2 Hybridization patterns of human genome DNA and 1 Hybridization patterns of sheep genome DNA.6. we got clones of Gal-1,ALDH,cath-D,ANXA2.Conclusion:1. Through RAPD and FP analysis,we find there may be gene mutation,gene site mutation,fragments missing,and so on.At the same time, there may be gene intrusion of E.G,these same to proteomics'changes.2. The probes got by RAPD and cloning may be used on disease diagnosis in future.
