Blueberry fruits'have rich nutrient, higher nutritional value and nutritional health care. Blueberries will be the most development potential fruit trees within the scope of this century. However, because of its low coefficient of propagation, it can not be large-scale produced, tissue culture is an important way to solve this problem. In this experiment, the stem of blueberry cultivar'Blomidon'was used as explants, the effect of the basic medium, ZT concentrations, sugar kinds, sugar concentrations and pH value on the proliferation and differentiation of blueberry adventitious bud were studied, and root system was optimized. In addition, the effect of several physicochemical parameters on the proliferation and growth of blueberries were studied during multiplying largely blueberry using air-lift bioreactor process, and changes of sugar, pH and EC of the medium were also studied in the process of bioreactor culturing blueberry, which provided a theoretical basis for scale production of seedlings from the blueberry cultivar'Blomidon'.The main results were as follows:B5 medium was the most appropriate medium for adventitious bud proliferation of blueberry; differentiation number of adventitious bud was the most on the medium of B5+ZT 2.0 mg·L-1 from blueberry stems, differentiation'number of adventitious bud was 6.8/strain; 40g·L-1 of sucrose concentration was the best for differentiation of adventitious bud, and differentiation number of adventitious bud was 7.75/strain; the value of pH 5.4 was the most suitable for differentiation of adventitious buds, and differentiation number of adventitious bud was 11.5/strain. IBA 0.5 mg-L"1 was most suitable rooting for blueberry varieties'Blomidon'in the process of blueberry rooting culture, the rooting rate reached 92.3%. In addition, blueberry cultured in the reactor, the contact cultural method was most suitable for proliferation and differentiation of blueberry, the number of adventitious buds splited up to 36.4/strain. Inoculating 60 blueberry stem in 5 L reactor was the best inoculation density, when ventilation was 0.10 vvm and light intensity was 2400 Lux, the proliferation and differentiation of blueberries in the reactor was the most productive, reached respectively 38.6,40.6 and 42.5/strain, which was significantly better than the number of proliferation and differentiation on the solid medium.
