Probiotics is some viable organism and metabolite,which can beneficially affect the host animal by improving its intestinal microbial balance.The common Probiotics include:lactobacillus,bacillus,yeast fungus and so on. There is one of Clostridium butyricum, as it's significant efficacy in clinical and health care,have been the focus of researchers. Besides, the Clostridium butyricum can be used to fermentation forage and have positive influence on forage industry. So, in this dissertation, the cultivation and application of Clostridium butyricum were Studied.The main results are as fellows:(1) The number of living bacteria cells was used as evaluate index,The purpose of research was to find the status of Clostridium butyricum in three kind of material. The results showed bean dregs is the best substantia for Clostridium butyricum. The maximum number of living bacteria cells of Clostridium butyricum could reached as about 47×106 CFU/g when used bean dregs. Under the same condition, the maximum number of living bacteria cells were 39×106 CFU/g and 34×106CFU/g separately when use wheat bran and maizena.(2) Response surface design method (RSM) was used to study the solid state fermentation of Clostridium butyricum. The living bacteria cells was used as evaluate index. The main factors of medium composition were screened out by Plackett-Burman experiments. Four main factors namely (NH4)2SO4,MgSO4,maltose and liquid water content were selected from 10 factors. Then results of Box-Behnken experiments showed that The optimum culture conditions were confirmed as follows:1.5% (NH4)2SO4,0.02% MgSO4,0.8% maltose and 55% liquid water conten.Under the condition, the number of living bacteria cells were 76×106CFU/g. Similarly, Clostridium butyricum solid culture conditions were optimized by Box-Behnken experimental design. The optimum conditions were obtained as follows:fermentation time 24h, temperature 30℃,14% inoculation, Under this condition, the number of living bacteria cells could be maximum that is about 11×107CFU/g.(3) It have been found that Clostridium butylicum and Escherichia-coli have the Mutualism relationship confirmed by the method of Microbial co-culture. The results showed that, compared with the control group,the number of living bacteria cells raised 70% and 58%.Besides,it was found that Clostridium butylicum has strong antagonism against Escherichia-coli.But, the fermentation broth of Clostridium butylicum have no such antagonism, which support the result of shugui xie contrary to the others.(4) The results showed that Clostridium butyricum can improve the intestinal health of piglets, reduce diarrhea, promote animal growthing, improve efficiency of forage and save costs of producing by the research of feeding.
