H. macrophylla 'Coerulea' is an important ornamental plant in garden. Based on the effects of Al stress on physiological characteristics of tissue culturing seedlings of H. macrophylla'Coerulea', this thesis found that it had strong Al-resistance.Main findings and conclusions are as follows:1. Medium MS+6-BA1.0mg/L+NAA0.2mg/L is easy to induce the clustered shoots.The results of the experiment on solidification of medium with Al press showed that when the nutrient components, the sterilization temperature and sterilization time were consistent, the higher Al concentration, the worse coagulum in the same pH and AGAR quantity; the higher pH,the better coagulum in the same Al concentration and AGAR quantity; the more AGAR quantity,the better coagulum in the same Al concentration and pH.2. Along with the increase of Al concentration and time, the Tissue Culturing Seedlings had an obviously different size. Seedlings in 0.25-1mmol/L were bigger and more exuberant than CK. The size of the Tissue Culturing Seedlings in 3-5 mmol/L were rarely changed against the seedlings before with little roots and whiten or amaranth leaves, even wither death.This thesis totally measured and analysed eight physiological indexes, including superoxide dismutase (SOD) activity, peroxidase (POD) activity, soluble Protein content, malondialdehyde (MDA) content, chlorophyll (Chl) content, soluble sugar content (SS), cell membrane permeability (IL) and free proline (Pro) content, etc. The results showed that SOD activity, soluble protein content and chlorophyll content are negatively correlated with Al concentration,the rest positively correlated. Thereinto the content of soluble sugar, cell membrane permeability and proline content are significantly positively correlated with Al concentration.Comprehensively, the Tissue Culturing Seedlings of H. macrophylla'Coerulea' had a tolerance of A1 with 2mmol/L, and growed better in low Al concentration.3.PBS solution and electrode buffer were used to extract protein. Based on SDS-PAGE,The author analysed the difference of proteins in the tissue culturing seedlings of H. macrophylla 'Coerulea' under different Al concentration. The results showed that both extraction can effectively extract protein, but electrode buffer extracted a more protein, strip more clearly. And the protein strips between the stress concentrations didn't have obvious difference.
