| Trichinosis is one of the most hazardous parasitical zoonosis, harming human health seriously, and severe case of which mainly died of myocarditis, cachexia and toxaemia. Therefore, It will be so important for the prevention of Trichinella spiralis and the development of new antiparasitic that the research on the pathogenic mechanisms of trichinosis. This study focused on the dynamic detection that Leptin,MIF and correlation of inflammatory factor in Trichinella spiralis- infected mouse, to demonstrate the mechanism of myocardium injury of Trichinella spiralis- infected mouse .According to the published sequence in GenBank, such as Leptin,MIF, Caspase9, Bax,Aapfa-1,PKB andβ-actin, were designed and synthesizeed primers. The total RNA of myocardium was extracted using Trizol method and then the objective genes were amplificated and assayed by semiquantitative RT-PCR means. The results showed that the expression ofβ-actin mRNA was fairly stable and high, but there is no significant changes; The relatve expression of Leptin mRNA was upturned, and reaching peak at 19~24 days p.i., then decreased at 42 p.i.; The relative expression changes of Bax mRNA,Apaf-1 mRNA,Caspase9 mRNA and PKB mRNA is similar to Leptin mRNA. The result presumed that Leptin and TNF-α, then enhance each other ,leaded to apoptosis, induced myocardium injury. The study detected that the expression of Bax mRNA,Apaf-1 mRNA,Caspase9 mRNA and PKB mRNAwere increased following the expression of Leptin mRNA, which showed that Leptin regulated inflammation reaction and inhibited apoptosis in myocardium,by mitochondria apoptosis. The concentration of intramuscular of dexamethasone on the myocardium injury of Trichinella spiralis- infected mouse.Which is good for control the expression of correlation factor by myocardium injury.It means dexamethasone can control the apoptosis of related gene and sauvegarde myocardique.Our concentration detected the metabolism changes of anti oxidative index consist of T-Sod,NO and ATP(Na+-K+-ATP),it on process of Trichinella spiralis- infected mouse. through it is compared with treatment group of dexamethasone that in the way of anti-inflammatory and anti-oxidant.Results show that the expression of the content of T-SOD had slight increase in one touch play at 9 days p.i post-infection, This was followed by an obvious increase at 14 days p.i.and decreased later. The content of NO reach a peak at 19 days, ATP content is apparent in the 9 day decreased trend, and continuing low dose of expression, 29 day later recovered slightly during. Dexamethasone treatment groups are similar with the infection. But all expressions are not the amount and infection set.Our concentration detected the hormons related to myocardial energy metabolism that consist of the content of .PPAR-α,AngII and insulin,it on process of Trichinella spiralis- infected mouse. through it is compared with treatment group of dexamethasone that in the negative accommodation way of lipid metabolism .Results show that AngII,PPARαand insulin have a similar tendency,that the content of them reach a peak at 19 ~24days,and following decreased later. Treatment group of dexamethasone had a similar tendency. But each of the expression all decreased,and inhibiting effect obviously.This experiment was conducted to study Leptin and MIF induced myocardial apoptosis in Trichinella spiralis-infected mouse, confirmed that Leptin and MIF induced cardiomyocyte apoptosis and caused metabolic disturbance of lipids, resulted in myocardium injury in the course of Trichinella spiralis infection.It revealed multiprotection design of glucocorticoid which inflammatory reaction and tissue damage ,though it is compared with treatment group of dexamethasone.But also will give instructive significance to lucubrate on pathogenic mechanism of Trichinella spiralis. |
