Bovine viral diarrhea virus is belonging to Flaviridae, Pestivirus genus (Pestivirus). Infections with BVDV cause heavy economic loss for both dairy and beef industries worldwide due to abortions, production of weak calves, congenital deformities, respiratory infections, thrombocytopenia, persistent infections (PI) and mucosal disease.During the infection, CD4 + T cells of the immune system mainly recognize two proteins NS3 and E2, which can induce immune system cells to produce antibodies against the protein. Therefore, detection of NS3 protein antibodies was considered as an effective way of detection of bovine viral diarrhea virus infection.In this study, both the recombinant NS3 protein and standard positive serum were employed to screen NS3 protein B cell linear epitopes, 6 reactogenicity protein peptides; (1VCKKITEHERCHVNI15),(20AFFGVMPRGTTPRAPVR36),(46RRGLETGWAYTHQGGI61),(281EGDMATGITYASYGYFC297),(426YSGEDPANLRVVTSQSPYVVVATNAIESGV455) and (481FIVTGLKRMAVTVGEQA497) were selected.The identification of B cell epitopes is important for understanding the antigenic structure, virus-antibody interactions at the molecular level, and is also useful in vaccine design and clinical application. Epitope selected not only for detection of BVDV antigen detection of serological testing to establish a variety of means, also in the future to carry out non-structural protein NS3 antigen or pathogenic mechanism of the structure provides a theoretical basis.Meanwhile, this study cloned the NS3 gene from bovine viral diarrhea virus (BVDV) VEDEVAC strain. The result showed that the average P-distance of Pestivirus NS3 amino acid sequence was 0.07 and the VEDEVAC strain was classified to BVDV type 1. |
