Animal oocytes, vector of maternal genetic information; supply materials of the embryonic development. It's also the driver and central regulator in the process of the follicular growth and development·The development condition and phenotypic variation of oocyte regulate sexual maturation, ovulation, early embryonic development and other key physiological processes of reproduction.Compared to the oocytes of other mammalian species, porcine oocytes have much more lipid in the cytoplasm. This study focuses on this specific phenotype and, by specific staining on liquid with Sudan III and oil red O, finds that a large number of lipid droplets are dispersed in the cytoplasm in such a way that they almost fill the the whole cytoplasm up, making it impossible to distinguish the cytoplasm from the nucleus and organelles.Transcriptome profile of porcine oocyte-cumulus cellscomplex(COCs) were examined by Agilent microarray and lipid-related genes werescreeningout to compared with that of other eight adipose tissue. Among the 1820 lipid-related gene transcripts found in swine,1561 were expressedin oocytes, According to the expression level, these gene can be grouped into four categories, high abundance expression (Cluster 1, the average expression 18.05), highly expressed (Cluster 2, the average expression level of 14.43), medium abundance expression (Cluster 3, the average expression level of 9.96)and the expression of low abundance (Cluster 4, the average expression level of 5.82). Compared with the COCs, the numberofdifferentially expressed genes in upper layer of backfat is 620, in which 284 increased,336 down; inner layer of backfat number is 594, of which 297 increased,297 down; greater omentum number is 618, of which 323 increased,295 down; lesser omentum number is 617, of which 313 increased,304 down; retroperitoneal adipose number is 615, of which 319 increased,296 decreased; abdominal fat number is 597, Of which 300 up-regulated,297 down; intermuscular fat number is 549, of which 334 increased,315 down; pericardial fat number is 493, of which 292 increased,201 decreased. These genes mainly participate in lipid metabolism process, fatty acid synthesis and metabolism process, cellular lipid metabolism process, lipid binding and other functional classification. In addition,21 oocytes-specificgenes were found expressed specific in the COCs, in which 12 up-regulationand 9 down-regulation. These gene distributed in nine cell lipid-related Go term. It is suggested that these genes may be related to the lipid regulation of pig oocytes and used as the candidate genes ofoocyte-lipid regulation, which is important for further study. These results help to understand the molecular mechanism of high lipid content of pig oocytes and provide an important basis data for the unique model of lipid deposition and fat metabolism in pig oocyte.
