It also provide a basis for further research and development toanthraquinones.The experimental subjects discuss the antibacterial and antitumor aspects ofbiological activity for14kinds of anthraquinone compounds in7kinds of commonpathogenic bacteria,3kinds of common pathogenic fungi and human hepatocellularcarcinoma cell Bel-7402by the drug sensitive paper sheet method, MTT stainingmethod, Annexin V and PI double staining method and PI single staining method.Andwith ceftriaxone sodium, fluconazole injection and doxorubicin hydrochloride (ADM)as the positive control medicine for efficacy analysis respectively, screening the activityof anthraquinone compounds for the test strains and the neoplastic cells, and analyzingthe strength of the activity for the effective anthraquinone compounds.Experimental results:Drug sensitive paper sheet method experiment selecte foursample concentrations, respectively0.5mg/mL,1.0mg/mL,1.5mg/mL and2.0mg/mL.They screen the antibacterial activity of anthraquinone compounds for the bacteria/fungi,and speculate the strength of the effective sample (Dihydrotanshinone I, LG02,Tripterine), the concentration range of minimum antibacterial concentration (MIC) andbest antibacterial concentrations (BAC) by a comparative analysis of the positivecontrol medicine. The antitumor biological activity research include MTT stainingmethod, Annexin V and PI double staining method and PI single staining method.Thesamples selecte the tripterine, Cryptotanshinone,Dihydrotanshinone I, Tanshinone I,Tanshinone IIA, Isobutyryl shikonin, Chrysophanol and Physcion for research.Thesample concentrations of MTT staining method experiment selecte1μg/mL,5μg/mL,10μg/mL and30μg/mL respectively.Results show that four medicine samples oftripterine, Dihydrotanshinone I, Tanshinone IIA and Isobutyryl shikonin have antitumorbioactive effects on human liver cancer cells Bel-7402. The concentration of drugs is30μg/mL in the Annexin V and PI double staining method. The result of antitumor effect show that tripterine>Isobutyryl shikonin>ADM>Tanshinone IIA>Dihydrotanshinone Iby analysising the cell apoptosis rate.The anticancer mechanism may be the inhibitionof cell proliferation in order to achieveing the purpose of apoptosis of tumor cells by PIsingle staining experiments. |
