| In this paper,Mortierella alpina 200012201,88335 and 87649 were studied with PDA plates and SEM observation for the first time.And we obtained the metabolic changes in ARA production by determining five value of three consecutive lots 20101128,20101206 and 20101213 in the production of Arachidonic Acid by Mortierella alpina.Furthermore,the orthogonal design of the medium composition in the production of Arachidonic Acid by Mortierella alpina was conducted,as well as the ARA oils were purified.All of these works laid the foundation for M.alpina 200012201 applied in the ARA industrial production.The results of SEM observation:the hyphae of the ARA high-yielding strain 200012201 were colorless,and the hypha branches were luxuriant,while few spores were found and only a small quantity zygospores were detected; The wild-type strain 88335 and 87649 produced more conidia which were oval or elliptical and bunchy,and no zygospores were founding.We infer that there is a certain relationship between ARA yield and M.alpina spores production:the ARA yield of M. alpina 200012201 with rarely spores production is higher,while M.alpina 88335 and 87649 which produced more spores rarely produce oil.The results of five values in three consecutive lots:the biomass (dry weight) was 30.650 g(/L zymotic fluid),sugar consumption was 117.560 g(/L zymotic fluid),nitrogen consumption was 0.0662 g/(100mL zymotic fluid),fat content was 17.142 g/(L zymotic fluid) and arachidonic acid content was 7.482 g/(L zymotic fluid).And these results compared to domestic similar studies are in great advantage,and the utilization rate still has ascend space.Orthogonal test was initially used to determine the fermentation medium for M.alpina 200012201.Glucose 80 g/L,corn flour 15 g/L,soybean meal 10 g/L,yeast extract 1.5 g/L,KH2PO4 1 g/L,MgSO4 0.1 g/L,pH 6.5.On this basis,a batch of fermentation,biomass reached 41.40 g/L,fat content was 17.54 g/L,and ARA content was 8.03 g/L.Using acetone as a solvent was better in low temperature crystallization for ARA oil purification,and ARA concentration was 60.5%,20% higher.And the mixed fatty acids were used as the object with methanol as a solvent,70℃for urea inclusion in ARA oil purification,and ARA concentration was 74.7%,about 35% higher. |
