Chapter 1:The physical and chemical properties, development and clinical research of anthracycline drugs were briefly summaried. And the dynamics of analiysis determination of anthracycline domestic and international were also described. The application of electrochememical determination in anthracycline drugs was mainly introduced. What is the more, the significance, contents and innovation of the reseach were introduced.Chapter 2:This paper used the linear sweep technique to study the electrochemical behavior of epirubicin hydrochloride. The results showed that there was a sensitive reduction peak at -0.34V (vs. SCE) in pH 6.80 Tris-HCl buffer. When the Bovine serum albumin was added, the value of the reduction peak was reduced deeply. Accordingly, a new electrchemical determintation method of bovine serum albumin was established. Under the optimum conditions, the detection limit of the epirubicin hydrochloride was 5.73×10-9mol·L-1. The decrease of the reduction peak currents of epirubicin hydrochloride were proportional to the concentration of bovine serum albumin in the range of 1.0×10-9~1.0×10-6mol·L-1 (R=0.9973). The detection limit of the bovine serum albumin was 8.05×10-10mol·L-1. And the binding constantβof the epirubicin hydrochloride and bovine serum albumin was 3.04×106L·mol-1 with the ratio was 1.Chapter 3:In this paper, the interaction between epirubicin hydrochloride and bovine serum albumin was investigated by fluorescence spectra. The experiment demonstrated that the fluorescence intensity of epirubicin hydrochloride was linear with its concentration in the range of 2.0×10-7 to 1.3×10-5mol·L-1, and its detection limit was 6.92×10-8mol·L-1. Moreover, the fluorescence intensity of epirbicin hydrochloride was enhanced when the BSA was added in. In the range of 3.0×10-8 to 2.5×10-7mol·L-1,the increase of fluorescence intensity (ΔF) was proportional to the concentration of bovine serum albumin. The detection limit of the bovine serum albumin was 5.05×10-9mol·L-1. Simultaneously, the effects of Fe3+, Ca2+ and Cu2+ to the binding of epirubicin hydrochloride with bovine serum albumin were also studied.Chapter 4:The chemical modification materials, such as grapheme were synthesized, and the bare glassy carbon electrode was modified. By the cyclic voltammetry method, the electrochemical behaviors of three anthracyclines on the modified electrode were investigated. And in the acidic buffer solution, the sensitivity of redox peak current of anthracycline drugs, such as doxorubicin, epirubicin and daunorubicin on graphene modified electrode was higher than the bare glassy carbon electrode. Choosing the experimental conditions, that is 60s accumulation time for the scan,-1.0V for the initial potential. And the problem how to eliminate the signal residue of the electrode was discussed. What is the more, the electrochemical behaviors of epirubicin on the grapheme modified electrodes were compared with its on the bare glassy carbon electrode. Also, the interaction between Bovine serum albumin and epirubicin was investigated. |
