Salicornia herbacea, a seasoned vegetable and traditional medicine, is commonly known as a halophyte that can grow in salt marshes, salt fields along the coastline. There are various of active constituents in S. Herbacea that are responsible for disorders such as constipation, obesity, cancer, anti-inflamatory, antioxidative effect and so on. This paper presents investigations on the water-soluble constituents of S.Herbacea and its antioxidative ability. The results were as follows:By comparison of four extraction methods of the water-soluble constituents in Salicomia herbacea including water soaking method, hot water soaking method, water-ethanol soaking method and ultrasonic assist water-ethanol soaking method, it is found that the antioxidant activity of the water-soluble constituents is highest when extracted by ultrasonic assist water-ethanol soaking method.The assist water-ethanol soaking method was selected to extract water-soluble constituents in Salicomia herbacea in this paper.The extraction condition was optimized and influence of the solid-liquid ratio, extraction time, extraction temperature and extraction times was studied. On the base of single factor experiments, It is selected the L9 (3~4) orthogonal table to go on the experiment with four factors and three levels. The results indicated that the best extraction condition was the conditions of 40℃,1:20 the ratio, 3 times and for 15min each time. Among these four factors,the most influencing factor is solid-liquid ratio, extraction times second, extraction time third and extraction temperature is the last.To study further the properties of the water-soluble constituents of S.Herbacea, separation and purification of the natural antioxidant was investigated. The experimental procedure consisted of the following three steps:liquid-liquid extraction, column chromatograph and high efficiency liquid chromatography(HPLC). Finally, two natural antioxidants were separated and identified.One is quercetin 3-O-β-D-glucopyranoside,and the other is 3-caffeoyl-4-dihydrocaffeoylquinic acid.
