Camphor tree is an evergreen plant, has antibacterial, has Qufengchushi, qi pain, blood circulation, Lee joints, and many other pharmacological activity, but also can absorb a variety of toxic gases and the fresh color protection and other effects. Currently, research has focused on the trees and plant cultivation, chemical composition analysis and active components of the functional research. In this paper, camphor tree leaves as raw material, of a camphor polyphenol extraction process, the final aqueous two-phase coupled with ultrasonic extraction methods and optimization of the Purification of polyphenol optimum camphor; Finally, studied more than camphor phenol hydroxyl radical, DPPH free radical clearance, clear simulated gastric juice nitrite, removing H2O2, reducing power and other aspects of the oxidation resistance and anti-lipid oxidation capacity.The main results are as follows:(1) Rotation through the quadratic regression combination design, by ultrasonic aqueous phase extracted two coupled mathematical simulation of camphor polyphenols equation:Where:X1 is the ultrasonic power, X2 for the solid to liquid ratio, X3 as extraction time, X4 for the extraction temperature.Determined by mathematical statistical analysis of ultra-aqueous two-phase ultrasonic coupling camphor polyphenols extracted optimal conditions: ultrasonic power is 254.7 w; solid to liquid ratio is 1:38.85; extraction time is 95.92 min; extraction temperature is 20.92℃, theoretical yield is 3.35% of polyphenols. Verification test and then found the optimum conditions, the polyphenol yield was 3.30%, process and reproducible model is reasonable(2) Purification using a large camphor tree polyphenols, the most suitable resin AB-8; AB-8 resin adsorption process in line with camphor polyphenols and Freundilch Langmuir equation, the adsorption process is endothermic, and the physical adsorption process; best on sample concentration 1.6 mg/mL, the sample solution pH= 5.68, the injection volume 2BV, elution volume 1BV, under these conditions, quadratic orthogonal rotation design optimization of the test sample flow rate, elution flow rate and eluent concentration, macroporous resin column chromatography mathematical model of camphor polyphenols:Where:X1 is the sample flow rate, X2 for the elution rate, X3 is the concentration of ethanol, after analysis:get Purification of camphor optimal conditions polyphenols, sample flow rate of 1.55 mL/min; elution flow rate of 1.72 mL/min; 63% ethanol, the purified camphor 76.79% yield of polyphenols.(3) Purification of three different samples of camphor polyphenol hydroxyl radical, hydrogen peroxide, nitrite and gastric juice have some scavenging DPPH, after Purification of the removal of the sample 3 is better; the restore the ability of the test substances experimental results show that the ability to sample 3, the reduction increases with the concentration, that is, within a certain range, the greater the concentration of the sample 3, the greater the antioxidant capacity. Camphor has a strong group of polyphenol samples Antioxidant capacity, the POV value of the relatively slow growth rate, and the higher the concentration, the stronger the antioxidant properties.(4) The results of structural identificationThis paper also camphor extracts were identified,what's more,the instruments and methods of chemical experiments, results showed that the extract contains camphor polyphenols; by gas chromatography-mass spectrometry identified further identified camphor extracts of betulin.In summary, ultrasonic aqueous two-phase extraction coupled camphor favor the extraction of polyphenols, a higher extraction rate. Camphor polyphenols scavenge hydroxyl radicals, hydrogen peroxide, DPPH radical and nitrite in gastric juice and other effects, can anti-lipid oxidation, can be developed into a natural antioxidant, to the camphor trees to provide more extensive use of resources means.
