Study On Extraction Process Of Supercritical Fluid Extract And Chemical Composition Of Non-volatile Material In Jinxuan Zhike XunxiSan

Jinxuan Zhike XunxiSan is a kind of Chinese medicine produced by Mayinglong Pharmaceutical Group Co., LTD. This preparation is composed of Flos Lonicerae Japonicae,Herba Schizonepetae,Herba Portulacae,Natrii Sulfas Exsiccatus,Calcine Alumen and so on. The preparation can clear away heat and toxic material, expel pathogenic wind,remove dampness, reduce swelling, ease pain, and curative effect is remarkable. To improve the extraction of Jinxuan Zhike XunxiSan, this paper have a study on the Supercritical Fluid extract which is the anti-inf lammatory's effective parts, including extraction craft, quality standard and fingerprint; To study the chemical constituents of Non-volatile material in Jinxuan Zhike XunxiSan.Using the supercritical CO2 extraction technology, this thesis conducts a study of the extraction craft of three Botanicals's supercritical CO2 extraction of Jinxuan Zhike XunxiSan through orthogonal design test, and got a reliable technical parameters, carried out the Medium-sized test validation, can interim to industrialization.This paper conducts a series of studies on the quality standard of supercritical CO2 extraction of Jinxuan Zhike XunxiSan, including traits,TLC,determination and so on. The results of this study provide a reference for improving its quality control standards.According to the "traditional Chinese medicine injections fingerprinting requirements (Interim) "which is issued by the state Food and Drug A dministration, established a fingerprint of the supercritical CO2 extraction in Jinxuan Zhike Xunxi San. Thirty-two peaks are selected as the common fingerprint peaks, they were clearly visible, have a good similarity and the determination method is reliable. Main component receive the adscription, which can response the relationship between number and quantity of the chemical composition of this extracts effectively.Using modern chromatographic separation and spectral identification method,9 monomer components were separated from the active parts which are purified by macroporous resin technology, 8 compounds were identified, they are Luteolin-4'-O-glucuronide butyl, Coffeic acid, Ethyl caffeate, Luteolin, Hesperidin, Luteoloside,5-0-caffeoyl-quinic acid,βValley Sterols. Among them, compound of Luteolin -4'-0-glucuronide butyl is a product of publicly reported for the first time, which is a new compound found in Mixed extracts. This is of great significance for the interpretation of material basis for efficacy of Jinxuan Zhike Xunxi San.
