| In recent years, Photodynamic Therapy developed a new type of tumor therapy, and the photosensitizers which play a decisive role in the treatment has become an active research field. As one kind of effective photosensitizer, phthalocyanine (Pc) and its derivatives have been considered to be a promising new kind of photosensitive drugs in PDT due to the clear structure and compose, easy control of quality, well photosensitive property and biology compatibility.In this paper, a series of liposolubility phthalocyanines with different structure have been synthesized, In addition, their spectrum property,photosensitive property,the mechanism of photochemical activity have been studied through the absorption and fluorescence spectroscopy and other means. Be used for the phthalocyanine photosensitizers, there is not a good solution to the physical environment of its solubility (ie water-soluble) and the aggregation problem, in fact, hindered the process used. Therefore an excellent exploration performance and stability in vivo transmission of water-soluble phthalocyanine photosensitizer delivery system become engaged in phthalocyanine phototherapy drug researchers to study. Which lay the basis for the application of Pc in the field of photodynamic therapy.The main research conclusions are as follows:(1) Three phthalonitrile derivatives(1a,2a,3a) and the corresponding phthalocyanines (4a,5a,6a), were designed and synthesized successfully, and were characterized by UV spectra, IR spectra and 1H-NMR. (2) By introducing of the ArO group, theλmax of metal zinc phthalocyanines redshift compared with none substituted phthalocyanines for certain degree. The fluorescence spectra of above compounds are similar, and have Stock shift about 13 nm. In addition, with the increase of aggregated phthalocyanines, the fluorescence intensity will decreases due to self-quenching.(3) The addition of surfactant can make phthalocyanine depolymerize, the non-ionic surfactants shows the better ability of dis-aggregation.(4) The results of the fluorescence spectroscopy showed that phthalocyanines (4a,5a,6a) have a powerful ability to quench protein BSA fluorescence, while changing the conformation of proteins.(5) In this paper, a new water-soluble phthalocyanine photosensitizer delivery system of phthalocyanine/silica nanoparticles (Pc-SN) was built by the micro-emulsion method. Compared with pc, Pc-SN had the singlet oxygen obvious increase in capacity. |
