Sample preparation plays an important position in analytical chemistry. Liquid-liquid extraction is a classic sample pretreatment method. This paper introduces a novel extraction technology—"sugaring-out" which is based on traditional liquid-liquid extraction and studies its application.Chapter 1:Several common sample preparation methods based on liquid-liquid extraction and their applications were reviewed and the principle and conditions of "sugaring-out" were summarized.Chapter 2:With addition of a monomeric sugar or a disaccharide into an acetonitrile-water solution, the acetonitrile would be separated from water to from a new phase. The NaDDTC complexes with Ni2-,Co2+,Cu2+ and Hg2+ were extracted into an acetonitrile phase from aqueous by adding glucose as a phase-separation agent and determined use HPLC. This study establish a new method for detection four metal ions in same time.Chapter 3:A new method for the simultaneous determination of five anthraquinone derivatives in Xiaoerresuqing oral solution is developed. HPLC on a Diamonsil C18 column (150x4.6 mm i.d.,5μm) is used, the mobile phase is methanol-0.1% phosphoric acid (85:15,v/v), at a flow rate of 1.0 mL/min, and the detection wavelength is 254nm. In the best condition, the five anthraquinones show good linear correlation in measuring range. The method is reliable for the determination of five anthraquinone derivatives.Chapter 4:A novel extraction method coupled with high performance liquid chromatography is established for the determination of 5-hydroxymethylfurfural. In this study, we investigate and optimize the volume of extraction solvent, extraction time, extraction temperature and pH value. The calibration curves of 5-HMF is linear in range of 1-50 g/mL. The method is simple and rapid for the determination of 5-HMF in sugary sample.
