Carnitine, Octopine are bioactive products which are very rich in octopus. Their nutritional and medicinal value has been widely recognized and its application prospect is broad. Production of high efficacious marine drugs and health food through the research is a effective way to achieve comprehensive utilization of the octopus resources.This thesis aimed to study the bioactive products-Carnitine and Octopine in octopus. A pre-column derivation RP HPLC method has been established to analyze Carnitine and Octopine. The HPLC method has an important significance on the assay of these compounds in aquatic product and the evaluation of food nutrition and security. The optimal conditions of the extraction of Cartinine and Octopine in octopus were determined by orthogonal experiment.1. A pre-column derivation RP-HPLC method was developed for the determination of Cartinine and Octopine with FMOC as the derivation agent. An Agilent HPLC system equipped with a Waters Symmetry C18 column (5μm, 150mm×4.6mm.ID) setting at a temperature of 35℃is used in this study. The mobile phase is composed of 0.6% triethylamine (pH 6.20) and methanol (40/60, v/v). The flow rate is 0.6 mL?min-1, the detection wavelength is at 263 nm. The most suitable derivatising condition are determined as follows:the pH of reaction buffer is 10.4; the concentration of FMOC is 30mmol/L; the temperature of reaction is 60℃and the time of reaction is 30min for Cartinine and Octopine.2. The acid extraction of Cartinine, Octopine from octopus was studied and analyzed by pre-column derivatization RP-HPLC. The optimal conditions were determined by orthogonal experiment as follows: With 5% TCA, ratios of material to solvent is1:4, ultrasound extracting temperature is 45℃, extracting time is 3 hours, the extraction is degreased by n-hexane and cleaned by chloroform/butanol (1:1, V/V). 3. The assaying of Cartinine and Octopine in octopus is preliminary studied in this thesis. The content of Cartinine and Octopine in different parts of octopus and in different months has some differences. And the month and season of harvest are influences on its content and can provide the basis for the development and utilization of octopus though future reseach.
