Lespedeza cuneata(Dum.Cours.)G.Don, also named Triofliumrepens or Mimosa pudica, mainly grow in hillside, waste land or on side of road. It is produced in the northeast of China, Jiangsu, Hunan and Yunnan. It contains many chemicals including pinitol, flavonoids, phenolic compounds, tannin andβ-sitosterol. D-pinitol (sennite) is an analogy of insulin. It can reduce blood glucose, promote absorption of creatine, cure disease which is caused by insulin resistance. It also has the function of relieving cough, expelling phelgm and typical anti-inflammation and anti-zcute edematous.To study extracting D-pinitol from Lespedeza cuneata(Dum. Cours)G.Don and choose the proper extraction solvent to extract D-pinitol from Lespedeza cuneata(Dum.Cours)G.Don, then separated and purified the extraction, finally characterized the purified.D-pinitol was analyzed by the high performance liquid chromatography-evaporative light-scattering detector(HPLC-ELSD) and showed good linear relationship in the range of 0.0346~0.173mg/mL with the equation:Y=4.9579X+5.8544 and linear correlation coefficient(r) was 0.9998, the relative standard deviation (RSD) was 1.73% with the add mark recovery rate 97.14%~103.31%.Through the solubility of D-pinitol in ethyl acetate, water and ethanols, water performed to be the suitable extractant. Five factors were considered to affect the rate of D-pinitol extraction in Lespedeza cuneata(Dum.Cours)G.Don, it included temperature, extraction time, solid to liquid ratio, alcohol concentration and ultrasound or not. The best condition to extract D-pinitol in Lespedeaza cuneata(Dum.Cours)G.Don was ultrasound in water with the ratio of liquid and materials for 1:18 under 95℃for 150 min. At last, the crude extracts of Lespedeaza cuneata(Dum.Cours)G.Don employed alcohol precipitation, then MS and NMR was used to character the precipitation. The HPLC-ELSD showed the content of D-pinitol in purification was 78.5%.
