Bio-membrane is a current active research area of molecular biology, cell biology. As one of the two components of membrane structure, membrane proteins play a key role in a variety of important basic biological processes, such as photosynthesis, respiration, nerve signaling, immune response and absorption of nutrients. Structure and function of membrane proteins is not only the key point in structural biology, biological chemistry and biological medicine, but also the crux of structure-dependent drug-designing. Studies have shown that integral membrane proteins account for30percent of total human proteins and proteins which interact with cell extima and intima50percent of total cell proteins. The versatility of membrane protein makes it an ideal drug target. Currently more than40percent of drugs around the world set membrane protein as target. Therefore, further studies of structure, folding and the its interaction with lipid will definitely contribute to elucidating function, developing highly-effective new drugs, preventing and curing diseases.Nuclear magnetic resonance is an interdiscipline to investigate molecular structure and dynamic conformation based on the interaction between nuclear spin and applied magnetic field. After several years'development in solid-state nuclear magnetic resonance (SSNMR) techniques, such as magic angle spinning (MAS), decoupling and recoupling cross polarization and so on, crowns SSNMR the unique superiority in researches of biological systems and makes SSNMR one of the powerful methods in protein structure, function and dynamics. The applicability of NOESY experiment has broadened from solution-state NMR to solid-state NMR thanks to the sample which processes a quick motion in liquid-gel phase and MAS technique. NOESY experiment can provide effective information of the space distances in proteins.In our project, we synthesized some amino acid methyl esters and peptides different in property and tried to clarify the interaction between the sample and lipid bilayers, conformation and location by the2D1H MAS NOESY experiments. Furthermore, the NOE buildup curve and the cross-relaxation rare were simulated initially. The preliminary study we've made in this work, will be the stepping-stone of peptide-lipid interaction model later.
