Indirect detection technology with high sensitive and high selective fluorescent probes for reactive oxygen species, particularly for singlet molecular oxygen, is currently of great importance. In this study, the photophysical and photochemical behavior of Singlet Oxygen Sensor Green (SOSG), a commercially available water-soluble fluorescent probe for singlet oxygen, was examined at first. The obtained results indicate that the fluorescence probe has high sensitive and selective detection for 1O2. In addition, thought SOSG and SOSG-EP is the efficient singlet oxygen photosensitizer, SOSG can be used effectively to detect the production of 1O2 with proper irradiation time. Second, in this study we use SOSG to study the 1O2 diffusion in single intact living cells. Our findings indicate that SOSG holds great promise for the indirect imaging of the 1O2 that can escape from single intact living cells. Depending on the site of'02 generation, 1O2 generated in the plasma membrane treated with PpIX can escape from the cell interior into the extracellular environment, while the 1O2 generated in the nucleus cannot.
