A strain which can grow optimally in culture media that contain 8%-10% NaCl was isolated from the Jilantai salt lake located in Inner Mongolia of China as a protease producers, Besides classic phenotypic characteristics which include morphological observation and the physiological and bio-chemical characteristics,16SrDNA analysis had been tesed.According to these,the strain could be placed in the genus Halobacillus as a novel speacies,for which we named Halobacillus sp.JLT-09,at the same time,it is a moderate halophile strain.It is rod shaped,Gram-negative,spore-forming,non-capsule-forming. Colonies are cream-coloured,smooth,moist,but edge is not very rules sleek.Optimal growth occurs in the presence of 8%-10%(w/v) total salt,in the presence of 3%(w/v)Mg2+,at pH8,at30℃-35℃.Mannitol,rhamnose,sucrose can be utilized as sole carbon,but the following compounds are not utilized as sole carbon:glucose,mannose,xylose,starch,lactose,galactose,ribose,sorbitol,arabinose,fructose. Methyl red and Voges-Proskauer are negative. Simmons′citrate test is negative,nitrate and nitrite are not reduced,H2S is not produced, it had non-amylase, urease lipase activity,but had cellulose enzyme activity and catalase is prodused.This strain production protease fermentation condition is optimized, and the results show that, by hydrolysis dairy protein for source of carbon and nitrogen source case, when the initial medium pH8; The incubation temperature for30℃; Inoculated quantity 0.5%, Training time 24h, medium of enzyme production conditions is the best. For the bacteria fermenting for cryogenic centrifugal, suction filter, ultrafiltration, lyophilization, get rough enzyme, the thick liquid enzymes fluid nature of the study, the results show that the optimal reaction pH for8- 10, the optimal temperature for 60℃role.
