Alternative pre-mRNA splicing is an important gene regulation mechanism for expanding proteomic diversity in higher eukaryotes. The misregulation of alternative splicing underlies many human diseases. Although the mechanisms of alternative splicing have been studied extensively, we had not begun to realize fully the diversity and complexity of alternative splicing regulation by an intricate protein-RNA network. Great progress has been made in genome-wide alternative splicing mechanism research, which provides a method and data support for construction of alternative splicing feature model.The main research goal is to integrate sequence features, establish genome-wide feature model of alternative splicing, analyze alternative splicing events and reveal the regulatory mechanism of alternative splicing.After the description of the biological mechanism of alternative splicing, the overall framework of research is shown, which provides a biological basis for analysis. Second, the systematic introduction of exon array design principles and work flow provides technical support for analysis. Third, based on methods of linear regression and information theory, a model of alternative splicing is established through the use of alternative splicing features and exon array. The final set includes 146 features. Fourth, compared with the previous methods, accuracy has increased when using this feature set to predict alternative splicing. Finally, though the analysis of AS2Disease database, the biological rationality of alternative splicing feature set is validated.In summary, alternative splicing feature model established in this paper can identify alternative splicing events effectively, which reveals the regulatory mechanism of alternative splicing.
