As a new potential class of antiviral drugs, antiviral protein has been a research focus in the world. It reported that housefly body contains antiviral substances. Housefly(Musca domestica L.) belong to Insecta, Diptera, Cyclorrhapha, Muscidae. In this study, we used housefly(Musca Domestica L.) larvae as the material to extract the housefly larvae crude protein, detected its antiviral and antitumor activity, the main results are follows:1. A de-lipoprotein has been extracted from the housefly larvae by Soxhlet extraction. Using Pseudorabies virus(PRV) as a target virus, chicken embryo fibroblasts (CEF) as a viral vector, studied the antiviral activity of the de-lipoprotein from the housefly larvae.Found that, the titer from the original 5.62×104 (TCID50 ml-1) decreased to 3.16×104 after the PRV and crude protein (0.1 mg/ml) were mixture at room temperature for 2 hours, indicate that the crude protein can inhibit the activity of the PRV; adding the crude protein 1 hour before infection and 1 hour after infection, the virus titers were 3.16×104 and 1.0×104, indicate that the crude protein may be sensitive at the phase of virus attachment or entry.2. A experimental was designed to research the influence of the crude protein to stage of PRV virus attachment and entry into CEF:adding the crude protein in land 2 hours before virus infection, and collect the supernatant after the viral infection cell culture 2 hours, virus titers were measured as 5.62×104 and 2.51×104, however, the supernatant contains large amounts of virus in either, it showed that added the crude protein before the virus infection, can affect the adsorption of PRV on CEF cells.3.Purified the crude protein by ion exchange chromatography, and two peaks were collected, Al and A2. Determine their antiviral activity, the titer of the PRV virus dropped from the 5.62×104 to 3.75×104 after mixed with purified protein A2(0.2mg/ml), purified protein Al had no effect on virus titer.4.MTT method was used to detect the anti-tumor activity of A1 and A2 protein on A549 tumor cells, The results show that A1 protein has no effect on A549 cells, but the A2 purified protein inhibited A549 cells significantly. And with the A2 protein concentration increased, the inhibition rate increased. A2 purified protein is the target protein that we need. In this study, We studied the anti-PRV virus and anti-tumor A549 cell activity of the protein from housefly larvae, And these results would be a base of further study of the insect anti-viral protein...
