Objective:To explore pisces(Tilapia) and Reptilia(Trionyx sinensis) islet cells separation, extraction method and function evaluation, provide a new donor source for clinical islet transplantation research.Methods:The separation of Tilapia and Trionyx sinensis islet cells by collagenase, to observe the morphological structure on light microscope; to detect cell survival rate with Trypan blue staining; Dithizone (DTZ) staining, Calculate the purity of isolated islets on morphological; used culture medium containing 10% fetal calf serum and RPMI 1640 to groom islet cell, stimulating islet cells using different concentration glucose in Vitro,and determining its bioactivity with ELISA.Results:Islet cells activity of Separation of Tilapia, Trionyx sinensis were more than 90%, islet purity in separation was more than 80%, Tilapia and Trionyx sinensis islet cells show definite biological response to different concentration glucose in Vitro, then increase of insulin yield according to increase of glucose concentration; Islet cells at the same time has the dose-effect relationship to two groups with different concentrations of glucose stimulated producing insulin(P<0.05), Groups at different times of the same concentrations of glucose stimulated insulin has no significant difference (P>0.05).Conclusion:The method of separation of Tilapia and Trionyx sinensis islet cells is feasible,the relation between insulin yield from Trionyx sinensis islet cells and different concentration glucose is positive correlation. Islet cells in the turtle under similar conditions tilapia insulin secretion from islet cells was significantly higher than Tilapia, this reflected the Reptilia pancreatic islets compared with Pisces is more advanced in biological evolution, which establish experimental basis for islet cell transplantation between reptilia and mammalia.
