Grimmia pilifera belongs to the Grimmiaceae Grimmia,it's a kind or drought tolerance moss,which plantcorpus are bottle green or dark purple and growing at the rock face or rock thin soil,in high mountains normally. Grimmia is important material to research drought response because it can withstand extreme drought and return to life as soon as be watered after drying a few years.This study base on full-length cDNA library of rehydrated Grimmia pilifera with switching mechanism at 5′end of RNA transcript (SMART) techniques,and single out one Cu/Zn-SOD gene and one eukaryotic translation initiation factor 4E gene,the former use Rapid Amplification of cDNA Ends technology to obtain the full-length of gene and the latter use in silico cloning technology.Have verified the two gene sequence,and analysed them by bioinformatics.The purpose are further revealed Grimmia molecular and functional theory in the process of drought tolerance,and establish theory base.Using RACE technology cloned GH120 gene which coding Cu/Zn-SOD. Sequence analysing indicate that it is 1288bp long and contains a integrity open reading frame which is 726bp long and code 241 amino acids ,with two functional sites. The gene encodes a protein is highly homologous with Physcomitrella patens,Picea sitchensis,Pinus sylvestnis var,Codonopsis lanceolata, and Sandersonia aurantiaca and etc,those who encode Cu/Zn-SOD,and is closest relationship with Physcomitrella patens.Using in silico cloning technology cloned GH425 gene which coding eukaryotic translation initiation factor 4E. Sequence analysing indicate that it is 726bp long and contains a integrity open reading frame which is 465bp long and code 154 amino acids ,with one functional sites. The gene encodes a protein is highly homologous with Physcomitrella patens,Picea sitchensis, Zea mays,Oryza sativa,Japonica Group and Arabidopsis lyrata and etc, those who encode eIF4E,and it has closest relationship with Physcomitrella patens,has closer relationship with Picea sitchensis.
