Optical Coherence Tomography (OCT) is a novel coherence-domain imaging technique, which is based on the exploration of short coherence length of broadband light sources to achieve non-invasive, real-time in vivo imaging. Due to its higher image acquisition, superior sensitivity and better signal-to-noise performance, Frequency domain optical coherence tomography (FD-OCT) has become one of the important topics in the biomedical optics areas.In this thesis both the imaging performance and the experimental realization of the FD-OCT were analyzed in detail. First of all, the history, development and current research status of OCT were introduced. Secondly, the basic theory and detailed arrangements as well as some important parameters of FD-OCT were discussed with the emphasis on the image reconstruction and signal processing. A whole Frequency domain OCT system was then designed and established, with which some preliminary experiments have been made. The experimental results were analyzed and the disadvantages of this system were pointed out. The result would be helpful for further research in FD-OCT.
