Optical Coherence Tomography can provide images of retinal structure which cannot be obtained by any other noninvasive diagnostic technique, so it has been the research focus of Optometry. Spectral Domain OCT expands the functions of OCT from single imaging in the time-region to pick up and analysis of spectral information. It has a broad application such as measuring the human retina structure.It is investigated on the structure and imaging principal of Spectral Domain OCT system. By using Fourier transform, the methods of moving reference mirror by a tiny position and phase shifting are analyzed. A method without reference mirror moving is discussed.The characteristic parameters of the elements of Spectral Domain OCT system are investigated and analyzed. A simulative optical experimental setup is built in Zemax prepared for dispersion compensation.Through theoretical analysis and numerical calculation, the dispersion of sample is discussed; the influence of group velocity dispersion on signal is calculated. Obviously, the resolution of OCT signals degrades without dispersion compensated. With the help of human eye model, hardware compensation and numerical compensation are discussed in human retinal imaging. Method combined hardware compensation with numerical compensation is researched. The simulative experiment results have shown this method is efficient.
