Objective: By exhaustion experiments on rats of sport ability, thus the grouping in rats. Detection of rat skeletal muscle cells HDAC5, GLUT4 protein expression levels and HDAC5 in the cell nucleus and histone H3 acetylation level. Thus research HDAC5 in the nuclei and the relationship between the GLUT4 protein expression.Method: Only 40 adult male wistar rats. Fitness swimming for three days, 30 min a day; the fourth day swimming exhaustion, and according to the records of each rat swimming time take top eight grades and final eight divided into congenital movement ability of high and low level in the two groups; 72 hours of rest, eliminate exhaustion after the effects of exercise on skeletal muscle, the materials. Of skeletal muscle cells and using western blot method of total protein HDAC5 and GLUT4 expression level and nucleoprotein HDAC5 and histone H3 acetylation level detection.Result:1, Sports ability high level group of sport ability is the average level of low level group was 2.47 times.2, Sports ability high levels of GLUT4 protein expression level of skeletal muscle cells is exercise capacity low level group was 2.96 times and the significant differences between two groups.3, Sports ability high levels of total protein in skeletal muscle cells HDAC5 protein expression level are exercise capacity low group was 0.42 times that in the two groups had no significant difference.4, Sports ability high level group of skeletal muscle protein in the nucleus HDAC5 expression level are exercise capacity low group was 0.093 times, with the significant difference between the two groups.5, Sports ability in the high level group of skeletal muscle nucleoprotein histone H3 acetylation level are exercise capacity low group was 5.785 times.6, In exercise capacity in high level group, 83% of HDAC5 finish out the nucleus, and in the sports ability is low level group, only about 25% of HDAC5 finish out of the nucleus.Conclusion:1, Sports ability of high and low level in the two groups of sports ability and its skeletal muscle cells of GLUT4 protein expression level is related to relationship, which is related to prove the relationship between the GLUT4 for glucose transfer function to the movement ability of decisive role.2, Movement ability of high and low level of the nuclei of two groups of HDAC5 GLUT4 expression level and the cell’s protein expression level of negative correlation relationship, proved that HDAC5 in prevent the transcription of GLUT4 protein in the nuclei. Sports ability level within the two groups of the nucleus histone H3 acetylation level and the expression of GLUT4 in cells is closely relative to levels of relationship, proved that HDAC5 in the nucleus content reduce, will weaken the histone acetylation, yes the space of chromatin structure is loose, DNA to stretch, exposed the GLUT4 transcriptional binding sites, complete transcription, raise the GLUT4 protein expression level.
