Behavioral sensitization was defined as an increased(behavioral) response to a given dose of drug or a response of a similar magnitude upon treatment with a lower dose of drug after repeated treatment with drugs of abuse.This experiment was conducted to establish the behavioral sensitization model,utilizing the animal model of stress induced by empty water bottles,and study:(1) Effects of stress on the expression of behavioral sensitization induced by morphine.(2) Effects of Huperzine A on the expression of morphine behavioral sensitization induced by stress.Then we discussed cholinergic system on drug dependence to supply science accordings for relapse therapy.We attempted to investigate effects of Huperzine A on the expression of morphine behavioral sensitization induced by stress.(l) Record the locomotor activities of rats in the development period.(2) Record the locomotor activities of rats in the transfer period.(3) Record the effects of stress on the expression of behavioral sensitization in the acute challenge of stress period.(4) Record the effects of stress on the expression of behavioral sensitization in the chronic challenge of stress period.The result of experiment suggested that the behavioral sensitization could be induced by repeated treatment with 3mg/kg morphine;the effects of environmental cue on rats which injected morphine would be reduced according to time;both acute and chronic challenge of stress could induce the expression of behavioral sensitization,which were reversed by administration of Huperzine A;after being injected morphine again,acute challenge of stress could not affect behavioral sensitization of the rats,prompting the power of stress was smaller than the reappearance of addictive drugs.The expression of behavioral sensitization induced by acute and chronic challenge of stress could be blocked by Huperzine A,suggesting that the cholinesterase inhibitor such as Huperzine A could be approached as potential therapeutic agents for patients who abuse drugs.
