0bjective:The purpose of this study was to investigate the changes of AQP4 and Kir4.1 expression to explore the mechanism of brain edema after CIRMethods:119 healthy male adult SD rats (250-320g) were divided into 2 groups randomly ,a CIR group and a sham operation control group。CIR model were established by occluding unilateral middle cerebral artery of the rats with suture method.the sutures were removed after 2 hours to allow reperfusion.The operation procedure in control group was the same as the CIR group except occluding unilateral MCA. The brains of the rats were taken out at 6h,12h,24h,48h,72h and 7d after reperfusion. The changes of BWC were measured by the wet and dry weight methods.Evans blue was used to measure the integrity of BBB.The expressions of AQP4 and Kir4.l were detected by immunofluorescence.The contents of AQP4 and Kir4.1 mRNA were detected by RT-PCR.The ratios of AQP4 mRNA and Kir4.1 mRNA were calculated by microsoft execel. Results :The brain water content of ischemic brain tissue increased form CIR 6h ,and reached its peak at CIR 48-72h. The permeability of BBB increased after CIR,the increasing permeability had two peaks,at CIR12h and CIR 48h , and the higher one was at CIR 48h .AQP4 and Kir4.1 were co-expressed at cerebral pia mater,choroid plexes,ependyma and perivascular .The expression of AQP4 increased after CIR and had one peak at CIR 72h,At same time, the expression of kir4.1mRNA was down during CIR 6-12h .Reperfusion after 12h, the expression of kir4.1mRNA begin to increase. The ratios of AQP4 mRNA and Kir4.1 mRNA were increase after reperfusion and had one peak at 24h.Conclusions:In the early stage after CIR,The increased capillary vessel permeability induced the brain edema,in the later stage.The redistribution of AQP4 and Kir4.1 played an important role in the pathological course of CIR brain edema. The brains of the rats were taken out at 1h,3h,6h,12h,24h,48h and 72h after reperfusion.HE staining was used to detect the pathological changes,The expression of AQP9 were detected by immunohistochemical.The contents of AQP9 mRNA were detected by RT-PCR .Results: AQP9 expressed at hippocampus,contex,circumventricular organs and osmorecepter after CIR.the expression of AQP9 mRNA and protein increased after CIR 1h and to the peak at CIR 12h.Conclusions In the early stage after CIR,The increased of AQP9 expression protected the brain ;In the later stage.The decreased of AQP9 was likely to correlated with the recovery of energy metabolism...
