Purpose: By studying colon and lung tissue SP-A mRNA of Constipation in rats expression changes and the impact of acupuncture on them, Elucidated the biological mechanisms of internal-external relation between lung and colon, provides the basis of internal-external relation between lung and colon for biology-based theory, further explore the control action of the acupuncture on internal-external relation between lung and colon disorders under pathology condition, provides the new mentality and the method for the research of acupuncture regulates related with zang-and fu-organs.Method: 48 Wista rats were randomly divided into 6 groups: blank group, capture group, model group, he-mu point group, the source point and the collateral point group, yuan-point group. The constipation model uses the medicine Compound Diphenoxylate given by intragastric administration method to make the model replicate; he-mu point group to take "Tianshu Point"with on"Shangjuxu Point"; the source point and the collateral point group to take "Hoku Point" with "Lieque point"; yuan-point group to take "Hoku Point"; the capture group captures every day, and fills the physiological saline to the stomach, does not carry on the acupuncture; the blank group feeds normally, does not carry on processing. Record each group's first bowel movement time after first time's administration, the collection and the record of administration latter 12 hours the excrement number of seeds and the excrement weight, and take 4 day-long accumulation quantity as the observations. After the experiment, extract the rats of colon, lung, small intestine, stomach, heart tissue and SP-A mRNA expression analysis of the differences of the organization of different groups Using RT-PCR technology.Result: 1 the first time bowel movement time of the rats, the model group and the capture group comparison has the significance difference(P<0.05), but the capture group and the blank group compares also has the significance difference(P<0.05); he-mu point group, the source point and the collateral point group compares with the model group has the significance difference(P<0.05), compare yuan-point group and model group, there is no significant difference(P>0.05); the source point and the collateral point group compares with he-mu point group, there is no significant difference(P>0.05), the source point and the collateral point group compares with the yuan-point group has the significance difference(P<0.05).2 Bowel movement number of seeds index in rats, the model group compares with the capture group has the significance difference(P<0.05), the capture group compares with the blank group also has the significance difference(P<0.05); he-mu point group, the source point and the collateral point group compares with the model group has the significance difference(P<0.05), compare yuan-point group and model group, there is no significant difference(P>0.05); the source point and the collateral point group compares with he-mu point group, there is no significant difference(P>0.05), the source point and the collateral point group compares with the yuan-point group has the significance difference(P<0.05).3 Bowel movement weight index in rats, the model group compares with the capture group has the significance difference(P<0.05), the capture group compares with the blank group also has the significance difference(P<0.05); he-mu point group, the source point and the collateral point group compares with the model group has the significance difference(P<0.05), compare yuan-point group and model group, there is no significant difference(P>0.05); the source point and the collateral point group compares with the he-mu point group has the significance difference(P<0.05), the source point and the collateral point group compares with the yuan-point group has the significance difference(P<0.05).4 Colon tissue and lung tissue of rats have SP-A mRNA expression, small intestine, stomach, heart tissue have no SP-A mRNA expression.5 SP-A mRNA expression of colon tissue, compare capture group and blank group, there is no significant differenc(eP>0.05); the expression has the elevation tendency, compare model group and capture group, there is no significant differenc(eP>0.05); the expression has the drop tendency, compare with model group, he-mu point group, the source point and the collateral point group ,yuan-point group have no significant difference(P>0.05); compare the source point and the collateral point group and he-mu point group, there is no significant difference(P>0.05); compare the source point and the collateral point group and the yuan-point group, there is no significant difference(P>0.05); the source point and the collateral point group compares with other acupuncture group, has the elevation tendency. Conclusion: 1 SP-A protein expression in the lungs and colon close ties between the protein or related substances, suggests that this theory of internal-external relation between lung and colon is correct.2 Acupuncture group of model with constipation improvement effect in varying degrees, he-mu point group and the source point and the collateral point group of model with constipation treatment effect in varying degrees, and the source point and the collateral point group surpasses yuan-point group. he-mu point group surpasses yuan-point group.
