Objective To investigate the expression of calpain 6 in cervical intraepithelial neoplasia (CIN) and cervical carcinoma, and to analyze its relationship with caspase-3 and microvessel density (MVD).Methods Paraffin-embedded tissues from normal cervical epithelium(n=10), CIN(n=30) and cervical cancer(n=65) were used to study calpain 6 and caspase-3 expression by immunochemistry. MVD in cervical cancer was labeled using anti-CD34 antibody.Results 1. The positive ratio of calpain 6 in the normal cervix, CIN and cervical cancer were 0.00%,43.33%,81.54%, respectively (H=30.05, P<0.01). Caspase-3 positive rate were 90.00%,43.33%,23.08%(H=13.24, P<0.05). MVD count were 9.50±3.21, 15.83±3.67,22.71±5.44 and the count gradually increased (F=25.77, P<0.01).2. The positive rate of calpain 6 in clinical stageⅠ,ⅡandⅢcervical cancer were increased gradually (χ2=7.840, P<0.05). Poorly differentiated group was higher than in middle-differentiated group (χ2=3.874, P<0.05) and in well-differentiated group (χ2=5.666, P<0.05), but highly differentiated group no significant difference with the middle group (χ2=0.182, P>0.05). The positive rate of calpain 6 with lymph node metastasis was higher than without lymph node metastasis (χ2=4.068, P<0.05).3. The expression of caspase-3 was not related to histological type, clinical stage and lymph node metastasis (P>0.05), but only to independent (P<0.05).4. MVD count of early stage was significantly higher than the later stages (t=3.674, P<0.01). Discrepancy between groups with different differentiation were significant (P<0.01). The differences between adenocarcinoma than squamous cell cervical cancer are significant (t=2.855, P<0.01). The MVD count with lymph node metastasis were significantly higher than without lymph node metastasis (t=5.949, P<0.01).5. The expression of calpain 6 was negatively correlated with caspase-3 (r=-0.543, P<0.01) and was positively correlated with MVD (r=0.977, P<0.01).Conclusions 1. The calpain 6 expression increased in cervical cancer and was correlated with the clinical stage, histological grade and lymph node metastasis. The calpain 6 may promote the development, invasion, metastasis and prognosis of cervical cancer.2. The expression of calpain 6 in cervical cancer was negatively correlated with caspase-3, indicating calpain 6 may be inhibits apoptosis and promotes tumor development by reducing the expression of caspase-3.3. The expression of calpain 6 in cervical carcinoma was positively correlated with MVD count, indicating calpain 6 may promote cervical cancer invasion and metastasis through the support of tumor angiogenesis.
