1-deoxynojirimycin (DNJ) is a kind of polyhydroxy piperidine alkaloid. Due to inhibitingα-glucosidase enzyme activity, it can inhibit postprandial blood sugar level and has some biological activities such as anti-virus and anti-tumor. DNJ is abundant in sericultural resources. In order to better utilize of DNJ, this paper focused on four DNJ extraction methods and investigated its distribution as well as influencing factors of DNJ biosynthesis in sericultural resources. Anti-tumor bioactivities were detected after DNJ was isolated and purified in the end. We anticipated the results can provide basic data and theoretical guidance to large-scale application of DNJ in sericultural resources.According to the literatures, among these four commonly used extraction methods: aqueous HCl immersion, aqueous ethanol immersion, decocting and microwave-assisted water immersion, aqueous HCl immersion extraction had the highest efficiency. However, the three others'efficiencies were only about 60% -70%. The results provided an important theoretical basis to determine efficiently extraction methods.Distribution pattern of DNJ in mulberry was analyzed by pre-column derivatization RP-HPLC after extracted with aqueous HCl immersion. The results found the content of DNJ vary in different tissues of mulberry. In upper leaves, it was between 0.3%-0.4%, in the lateral root 0.2%-0.3%, in the middle part of leaves 0.1% -0.2% and in the low part leaves less than 0.1%. It indicated DNJ was rich in young leaves of mulberry trees.We also studied DNJ in Husang-32 leaves from different geographic regions. The results showed that DNJ contents differed in different locations. The contents ranged from 0.06% to 0.21% while latitudes, pH and contents of mineral elements in relative soil were also different. Moreover, DNJ had two synthesis peaks during 6-10 a.m. and 6 p.m. every day. These results suggested that biosynthesis of DNJ not only was connected with its own metabolism, but also some other factors such as geographical location, soil properties, meteorological and environmental factors.Drying and pulverizing were two important pre-treatments of DNJ extraction from silkworm powder. Studies indicated freeze-drying was better than hot-air drying. It was more efficient from smaller particle size during DNJ extraction. After aqueous HCl immersion, DNJ was isolated and purified by 732 strong acid cation exchange resins. DNJ of SCS (component separated from silkworm) can reach 1.08%. Based on the results of MTT test, SCS had significant anti-tumor activity. The inhibition of SCS to tumor cells showed a remarkable dose-dependent manner.
