Objective:To study the the dynamic expressions and their significances of Tumour Necrosis Factor-aand the typeⅠand typeⅡreceptor for TNF-ain mice with lung fibrosisMethod:we wil use the following research methods ,Ninety male healthy wister mice were randomly divided into three groups.The first group is the control group.It has thirty mice.The second group which has thirty mice is the disease model group .The third group which has thirty mice is the Prednisolone-treated group.,control group was poured by saline, the disease model group and the Prednisolone-treated group were poured by bleomycin solutions (5mg.kg-1) and the Prednisolone-treated group were injected in abdomen with Prednisolone(15mg/kg) in the same day,once everyday.The control group and the disease model group were also injected in abdomen with saline (0.3ml/只), once everyday. getting blood from the abdomimal aorta on the 1st,3rd,7th,14th,21th and 28th day than killed them. Collecting the bronchoalveolar lavage fluid(BALF)and the lung tissues.ELASA method was used to analyse the content of TNF-ain BALF and the contents of TNF-α,TNFR1andTNFR2 in bloodserum,immunohischemistry method was used to analyse the expressions of TNFR1,TNFR2 in the lung tissues .Base water resolution method was used to analyse the expressions of hydroxproline in lung tissues. Taking some datas and making some statistics analysis in the end.Result: The first group(control group)The cotent of TNF-αin bronchoalveolar lavage fluid(BLAF) and the content of TNF-α,TNFR1,TNFR2 in blood serum were a little low.The expressions of TNFR1,TNFR2in lung tissues were also weak.The content of hydroxproline in lung tissues was also low.Histopathological examination expressed lung tissue structure were normal .The white blood cell total and the percentage of neutral granalar cell in bronchoalveolar lavage fluid in the different period had no significantly change.The second group(the disease model group) The contents of TNF-ain BALF and blood serum were higher than those in control group at every time points.They have statistics differences(P<0.05).The content of TNF-ain BALF and blood serum increased obviously on the 7th day and peaked on the 14th day, then deceasedl.The contens of TNFR1,TNFR2 in blood serum increased with the time and peaked on the 21th day.then decreased.The contents of both the receptors were still higher than those in the control group on the 28th day.The contents of both the receptors in the disease model group have statistics differences when compraring with the control group at every time points(P<0.05).The protein expressions of TNFR1,TNFR2 in lung tissues were strong and peaked on the 21th day.The expressions of TNFR1,TNFR2 in lung tissues have statistics differences when compraring with the control group at every timepoints(P<0.05).The content of hydroxproline in lung tissues increassed with the time and higher than it in the control group at every time points.They have statistics differences(P<0.05).The inflamationg cells increased obviously on the 3 th and 7th day in histopathological examination.Alveolar intermission changed a little thick and the alveolar space changed a little smaller on the 7th day.Alveolar intermission changed more thick obviously and alveolar space changed more smaller obviously.Fibroblasts increased.The lung tissues were destroyed and fibrosis were more obviously on the 28th day.The white cell total and the percentage of the neutral granalar were higher than those in the control group at every time points. They have statistics differences (P<0.05).Then they peaked on the 7th day and the 14th day.The third group(the prednislone treated group) the content of TNF-ain BALF and the contents of TNF-α,TNFR1,TNFR2 increased a little at low level,they were lower than those in the disease model group and decreased obviously from the 7th day,they have statistics diffferences (P<0.05).The content of them were still higher than those in the control group, they have statistics difference (P<0.05).The protein expressions of TNFR1,TNFR2 in lung tissues were a little weaker when comparing with the disease model group and they were more weaker on the 7th day,they have statistics difference (P<0.05).But the protein expressions of both the receptors were still higher than those in the control group at every time points,they have statistics difference (P<0.05).The content of hydroxproline in this group was a little lower when comparing with the disease group and dicreased more obviously on the 7th day.They have statistics difference (P<0.05).but the content of hydroxproline were still higher than it in the control group at every time points and they have statistics difference (P<0.05).Alveolar structure was a little informal in histopathological examination.A little inflamation cells could be seen on the 3rd or 7th day. Alveolar space wasn't destroyed on the 14th day or 28th day.The white cells total and the percentage of nertral granalar increased at low level,they were a little lower than those in the disease model group,they have statistics differenc(P<0.05).but still higher than those in the control group at every time points and they have statistics difference(P<0.05).Conclusion:①TNF-αwill has an important role in the progression of lung fibrosis through the signal transduction ways of TNFR1and TNFR2.. It can also conclude that The PMN of TNF-a protein expression masculine in alveolar macrophages markedly increased in acute inflammatory so it play an important role in Bleomycin-induced pulmonary fibrosis.②.It concludes that Prednislone resisted its inflammatory reflection.It indicated that prednislone was the valid protection and treatment drug in the pulmonary injury through its resisted its inflammatory reflection.③.TNF-α,TNFR1,TNFR2 may be served as a guided index in clinical diagnosis and treatment of pulmonary fibrosis.
